Abstract:

Die pflanzliche Aquaporin-Familie der Nodulin 26-ähnlichen intrinsischen Proteine wurde aufgrund ihrer strukturellen und funktionellen Ähnlichkeit mit Nodulin 26 aus der Sojabohne benannt. Im Rahmen dieser Arbeit sollte das zu dieser Familie gehörende Arabidopsis thaliana Aquaporin AtNIP1;1 funktionell charakterisiert werden. Das besondere Interesse galt dabei einer möglichen Ammoniakleitfähigkeit dieses Aquaporins, da es eine hohe Sequenzähnlichkeit mit dem ammoniakleitfähigen Nodulin 26 besitzt. AtNIP1;1 wurde mit Hilfe eines Hefeexpressionssystem im Hinblick auf seine Wasser- und Ammoniakleitfähigkeit untersucht. Dabei zeigte sich, dass AtNIP1;1 eine moderate Wasserleitfähigkeit besitzt und zudem eine Ammoniakleitfähigkeit aufweist. Bei vielen Aquaporinen lässt sich die Wasserleitfähigkeit mit Hilfe von Quecksilberchlorid inhibieren. AtNIP1;1 wurde in der vorliegenden Arbeit auf seine Quecksilbersensitivität hin untersucht. Es konnte dabei zwar die Ammoniakleitfähigkeit von AtNIP1;1 durch Quecksilberchlorid inhibiert werden, nicht jedoch die Wasserleitfähigkeit. Dies wirft die Frage auf, ob die beiden Moleküle unterschiedliche „Wege“ durch das Aquaporin nehmen. Um Hinweise auf die Funktion des Aquaporins in der Pflanze zu erhalten, wurde die Expression und Lokalisation von AtNIP1;1 untersucht. AtNIP1;1 wird nahezu ausschließlich in der Wurzel von A. thaliana exprimiert, lediglich in den Samen findet sich ebenfalls eine erhöhte mRNA-Akkumulation. Eine Analyse der Promotor-Aktivität mittels einer AtNIP1;1-Promotor/GUS-Fusion zeigte eine Aktivität des Reportergens in der Wurzelspitze und im Zentralzylinder der Wurzel, sowie in den Leitgefäßen der Blätter. Zudem wurde die subzelluläre Lokalisation von AtNIP1;1 ermittelt. Dabei zeigte sich eine Lokalisation des Aquaporins in der Plasmamembran. Die Ergebnisse weisen auf eine Funktion von AtNIP1;1 in der Regulation des Wasser- und Stickstoffhaushaltes der Pflanze hin. Um Aussagen über die physiologische Funktion von AtNIP1;1 in der Pflanze treffen zu können, sollten Untersuchungen mit entsprechenden T-DNA-Insertionslinien durchgeführt werden. In diesen Linien lagen jedoch zusätzliche T-DNA-Insertionen in der genomischen DNA von A. thaliana vor, so dass diese zunächst ausgekreuzt werden müssen, ehe mit ihrer Hilfe Funktionsanalysen im homologen System vorgenommen werden können.

The plant aquaporin family of the Nodulin26-like intrinsic proteins was named because of the high structural and functional similarity to soybean Nodulin 26. The aim of the thesis was the functional characterization of the Arabidopsis thaliana NIP-aquaporin AtNIP1;1. The main focus was on a possible ammonia conductivity of this aquaporin as it shares high homology to the ammonia conducting Nodulin 26. In an yeast expression system AtNIP1;1 was in this work functionally characterized as water and ammonia conductive. For many Aquaporins the water conductivity exhibits mercury sensitivity. For AtNIP1;1 it was not possible to inhibit the water conductivity with mercury but the ammonia conductivity showed mercury sensitivity. This raises the question whether both substrates take the same passage through the aquaporin. In order to get hints about the function of AtNIP1;1 in planta, the expression pattern and the localization of AtNIP1;1 was investigated. AtNIP1;1 shows highest mRNA-accumulation within roots of A. thaliana. Furthermore there is also a higher mRNA-accumulation in the seeds, compared to the other parts of the plant. An analysis of the promoter activity with an AtNIP1;1-Promoter-GUS-fusion lead to reporter gene activity in the root tip and in the vascular bundles of roots and leaves. An investigation of the subcellular localization of AtNIP1;1 revealed that it is localized in the plasma membrane. The results of this work indicate a function of AtNIP1;1 in the water and nitrogen metabolism of A. thaliana. To find evidence for the physiological functions further investigations, for example with AtNIP1;1-T-DNA-insertion lines, should be performed.
