Abstract:

Kubisch-oktamere Silsesquioxane (“cube-octameric silsesquioxanes“, COSS) sind anorganisch-organische Hybridnanomoleküle mit einem würfelförmigen Si8O12-Kern und 8 organischen Gruppen, die ausgehend von den Silizium-Eckatomen in sämtliche Oktanten des kartesischen Raumes streben. Ausgehend von einem COSS mit 8 Azidgruppen wurden Glycocluster synthetisiert, die 8 bzw. 16 Kohlenhydrateinheiten auf ihrer Oberfläche präsentieren. Zum Aufbau der Glycocluster wurde die Kupfer-katalysierte Azid-Alkin-Cycloaddition (CuAAC) sowie Reaktionsfolgen von thermischer Azid-Alkin-Cycloaddition und CuAAC bzw. thermischer Azid-Alkin-Cycloaddition und radikalischer Thiol-En-Reaktion eingesetzt. Weiterhin wurde eine breit einsetzbare Direktsynthese für Acetylendicarboxamide entwickelt und mit dieser Methode eine Vielzahl von Acetylendicarboxamiden hergestellt. Das Reaktionsverhalten von Acetylendicarboxamiden in thermischen Cycloadditionen (Diels-Alder Reaktionen, dipolare Cycloadditionen) und Thiol-Michael-Additionen sowie die daraus resultierenden Anwendungsmöglichkeiten als Dendrimerbausteine, AB2-Bausteine, orthogonale AB2-Bausteine und als Reagenzien zur Proteinkonjugation wurden untersucht.

Cube-octameric silsesquioxanes (COSS) are inorganic-organic hybrid nanomolecules with a cubic silica-like core decorated with an organic group at each silicon apex. An octaazide functionalized COSS was decorated with carbohydrates resulting in glycoclusters bearing 8 respectively 16 carbohydrate units at their surface. Reactions to build up these glycoclusters were the copper-catalyzed azide-alkyne cycloaddition (CuAAC) as well as reaction sequences consisting of thermal azide-alkyne cycloaddition with CuAAC and thermal azide-alkyne cycloaddition with radical thiol-en reaction, respectively. Furthermore, a broadly applicable facile direct synthesis of acetylenedicarboxamides was developed, and numerous acetylenedicarboxamides were synthesized. The reaction behaviour of acetylenedicarboxamides in thermal cycloadditions (Diels-Alder reaction, dipolar cycloaddition) and thiol-Michael additions were investigated, as well as their possible applications as dendrimer building blocks, AB2-building blocks, orthogonal AB2-building blocks and suitability as reagents for protein conjugation.
