Abstract:

Die lichtabhängigen, vorübergehenden Oszillationen des Membranpotentials wurden mit Glas-Mikroelektroden in Mesophyllzellen höherer Pflanzen (Oenothera, Avena) gemessen. Die Oszillationen fehlen in Blättern von Mutanten der Nachtkerze Oenothera, die einen Genomdefekt im Photosystem I oder II haben. Ein intaktes Zusammenwirken beider Photosysteme ist offenbar notwendig für das Auslösen der lichtabhängigen Oszillationen. Oszillationen können auch in weißen, mutierten Zellen gemessen werden, sofern sie mit grünem Gewebe in Kontakt stehen. Daraus wird eine effektive symplasmatische Weiterleitung der Membranpotentialschwankungen abgeleitet, die durch elektronenmikroskopische Untersuchungen unterstützt wurde. Cycloheximid, ein Hemmstoff der Proteinsynthese, reduziert die Oszillationen erheblich, ohne dabei die Atmung oder Photosynthese zu beeinflussen. Das Ausbilden der lichtabhängigen Oszillationen ist auch mit dem Ergrünen von etiolierten Avena-Koleoptilen korreliert, wobei eine Korrelation mit dem lichtindizierten Protonentransport und der ATP-Konzentration vermutet wird.

Light-triggered transient oscillations of the membrane potential are recorded with glass micro-electrodes in mesophyll cells of Oenothera and Avena. These transients are absent in leaf samples prepared from variegated plastome mutants of Oenothera, in which all cells have a genetically impaired photosystem I or photosystem II. This suggests that co-operation between the two photosystems is responsible for the induction of the transients. Transients are observed in mutated cells when the illuminated leaf sample also comprises normally green cells and there is a symplasmatic connection between the two types of cells. This phenomenon of electric coupling between neighbouring cells is discussed in view of the concept of symplasmatic transport.
