Abstract:

Die beta-Sekretase und das Proteasom gehören zur Gruppe der Peptidasen. Während die beta-Sekretase eine Aspartylprotease ist, handelt es sich beim Proteasom um eine Threoninprotease. In einer Inhibition des jeweiligen Enzyms werden medizinische Anwendungen gesehen. Die Synthese der entsprechenden Inhibitoren basiert auf der Derivatisierung von N-geschützten Aminoalkoholen, ihrer entsprechenden Aldehyde oder verwandten Peptid-Derivaten. Die beta-Sekretase ist ein Target in der Behandlung der Alzheimerkrankheit. Indem sie das Protein APP (amyloid precursor protein) in dessen beta-Position spaltet, wird ein Protein C99 generiert, das durch die gamma-Sekretase zu Amyloid-beta-Peptiden (A-beta) prozessiert wird. A-beta kann Aggregate und extrazelluläre Plques bilden, die eine pathogene Kaskade einleiten und zu neuronaler Dysfunktion führen. beta-Sekretase-Inhibitoren können die Spaltung von APP durch die beta-Sekretase verhindern. Eine Anzahl von Inhibitoren wurde synthetisiert. Diese Inhibitoren basieren auf einem Isophthalamid und enthalten in der Mehrzahl ein Hydroxyethylamin-Übergangszustandsisoster. Zu ihrer Synthese wurde die Grignard-vermittelte Addition einer Iodmethyl-Gruppe an N-geschützte Aminoaldehyde untersucht. Die resultierenden, enantiomerenreinen sekundären Alkohole sind meist (S)-konfiguriert und besitzen daher eine zu allen anderen Hydroxyethylamin-basierten beta-Sekretase-Inhibitoren eine invertierte Konfiguration. Diese Inhibitoren zeigen keine oder eine schwache Aktivität im beta-Sekretse-Assay. Einige moderat aktive Inhibitoren wurden unter Verwendung der beta-Iodethanol-Intermediate hergestellt. Das Ubiqutin-Proteasom-System ist der hauptsächliche Weg des intrazellulären Proteinabbaus. Es spielt eine wichtige Rolle in zahlreichen zellulären Prozessen, was es zu einem Target in der Behandlung verschiedener Krankheiten macht. Das 26S-Proteasom in eukaryotischen Zellen ist ein Multienzymkomplex, der aus zwei 19S-Komplexen und dem 20S-Proteasom besteht. Das 20S-Proteasom enthält drei verschiedene, aktive Untereinheiten (beta1, beta2, beta5). Zur Synthese von Proteasom-Inhibitoren wurden Tripeptidalkohole und ihre entsprechenden Aldehyde in verschiedene funktionelle Gruppe umgesetzt. Die resultierenden Verbindungen wurden in verschiedenen Proteasom-Assays untersucht. Die aktivste Verbindung ist ein alpha-Ketoamid, das einen P1'-Rest enthält. Da die meisten Proteasom-Inhibitoren in Richtung der S-Taschen orientiert sind, stellt dieses alpha-Ketoamid einen Weg dar, die Eigenschaften der S1'-Tasche zu untersuchen. Die Tripeptid-Einheit basiert auf MG132 (Z-Leu-Leu-Leucinal). Die Inhibitoren zeigen eine gute Aktivität in zellfreien als auch im zellulären Assays.

The beta-Secretase and the 20 S proteasome belong to the group of peptidases. While the beta-Secretase is a member of the aspartic proteases the proteasome is a threonine protease. The inhibition of either enzyme may be used in medicinal applications. The synthesis of their inhibitors is based on the derivatisation of N-protected amino alcohols, their respective aldehydes or related peptide derivatives. The beta-Secretase is as a promising target in the treatment of Alzheimer's disease. Upon cleaving the protein APP (amyloid precursor protein) in its beta-site, the a protein C99 is generated which is processed by gamma-Secretase to the amyloid-beta-peptide (A-beta). A-beta can form aggregates and extracellular plaques that lead to the death of neurons via a pathogenic cascade. beta-Secretase inhibitors can prevent the pathogenic cleavage of APP. A number of potential inhibitors were synthesized. They are based on an isophthalic amide and most of these inhibitors contain a hydroxyethylamine transition state isostere. For their syntheses the Grignard-mediated addition of an iodomethyl group to N-protected amino aldehydes was investigated. The resulting, enantiomerically pure secondary alcohols in the inhibitors are mostly (S)-configured and thus possess the opposite configuration compared to known hydroxyethylamine beta-secretase inhibitors. These (S)-configured alcohols show no or weak activity in the beta-Secretase assay. Several other moderate inhibitors were generated using beta-iodoethanol intermediates. The ubiquitin-proteasome-system is the major pathway for intracellular protein degradation. Its implications in many cellular processes make it a target for the treatment numerous diseases. In eukaryotic cells, the 26 S proteasome is a multienzyme complex which consists of two 19 S complexes and the 20 S proteasome. The 20 S proteasome contains the three different active subunits (beta1, beta2, beta5). For the synthesis of proteasome inhibitors, tripeptide alcohols and their respective aldehydes were converted into different functional groups. The resulting compounds were investigated in different assays. Most promising is an alpha-ketoamide that contains a residue P1'. Most proteasome inhibitors are oriented in the direction of the S-subpockets. Thus, the alpha-ketoamides constitute a way to investigate the characteristics of the S1'-subpocket. The tripeptide backbone is based on MG132 (Z-Leu-Leu-Leucinal). The inhibitors display good activities in cell-free and cellular assays.
