Abstract:

Die Suche nach neuen Methoden zur effektiven, zielgerichteten Entwicklung neuer Wirkstoffe ist ein hochaktuelles Thema in der akademischen und industriellen Forschung.Ziel dieser Arbeit war es, die von Reggelin et al. entwickelte Methode zur asymmetrischen Synthese hochsubstituierter Azapolycyclen als flexiblen Zugang zu potentiellen Wirkstoffen und dabei in erster Linie zu nicht-peptidischen Peptid-Mimetika in Bezug auf strukturelle und methodische Aspekte auszubauen. Im Speziellen sollte dabei ein Zugang zu β-Turn-Mimetika geschaffen werden.Im Rahmen der hier vorgestellten Arbeit gelang es, alle vier möglichen Isomere eines Azabicyclus herzustellen und zwei von diesen mit pharmakophoren Gruppen zu dekorieren. Ziel war es dabei NK1-antagonistisch aktive Verbindungen zu synthetisieren und in einem funktionellen Assay auf ihre Wirkung hin zu testen. Darüber hinaus konnte das Indol-und Indolin-Strukturfragment als neuer Ringtyp in die Methode zur Synthese von hochsubstituierten Azapolycyclen integriert werden. Es wurde des weiteren gezeigt, dass die Synthese der „West-Seiten“-funktionalisierten 2-Alkenylsulfoximine und Azabicyclen in guten bis sehr guten Ausbeuten möglich ist. Durch die Synthese solcher funktionalisierten Azabicyclen konnte die Methode strukturell entscheidend erweitert werden. Die Entwicklung der ersten Methode zur funktionalisierenden Desulfurierung für Sulfoximin-substituierte Azapolycyclen war ebenfalls erfolgreich. Die auf der Umsetzung eines an seiner α-Position deprotonierten Azapolycyclus mit einem Carben beruhende Julia-artige Reaktion liefert die Vinyl-substituierten Azapolycyclen mit guten bis sehr guten Ausbeuten. Die neue Methode weist dabei außerdem eine sehr große konstitutionelle Breite auf. Es können verschiedene Ringgrößen, Substitutionsmuster, Stickstoff-Schutzgruppen und neben Azabicyclen auch Azapolycyclen vom chiralen Sulfoximin-Auxiliar funktionalisierend abgespalten werden.Durch die zahlreichen strukturellen und methodischen Fortschritte in dieser Arbeit besteht nun also die Möglichkeit die von Reggelin et al. entwickelte Methode zur asymmetrischen Synthese hochsubstituierter Azapolycyclen als flexiblen Zugang zu potentiellen neuen Wirkstoffen zu nutzen.

The search for new methods for the effective, target-oriented development of new active substances is an ongoing topic in the academic and industrial research. In this work a method for the asymmetrical synthesis of highly substituted azapolycycles which was developed by Reggelin et. al., was extended to a flexible entrance to potential new active substances and primarily to nonpeptide peptide-mimics regarding structural and methodical aspects. Of special interest was thereby an approach to β-turn-mimics.Within the scope of this work all four possible isomers of an azabicycle were synthesized and two of these were decorated with pharmacophoric groups. The pharmacologic goal were NK1-antagonistic active compounds which should be tested in a functional assay on their effect. Furthermore the indol- and indolin-structure fragment could be integrated as a new type of ring into the method for the synthesis of high-substituted azapolycycles. It was also shown that the synthesis of the “west-side“-functionalized 2-alkenylsulfoximins and azabicycles are possible in good to very good yields. The method could be crucially structurally extended by the synthesis of such functionalized azabicycles. The development of the first functionalizing desulfuration method of sulfoximin substituted azapolycycles was likewise successful. The method, based on a Julia-reaction-like conversion of an α-deprotonated sulfoximin with a carbene supplies vinyl-substituted azapolycycles with good to very good yields. The new method exhibits thereby in addition very large constitutional width, as different ring sizes, substitution samples and groups of nitrogen protection can be used.By the large number of structural and methodical improvements in this work it is now possible to use the method for the asymmetrical synthesis of high-substituted azapolycycles, introduced by Reggelin et al., as flexible entrance to potential new active substances.
