Abstract:

Das Griselimycin-Derivat Methylgriselimycin (MGM) zeigt sowohl in vitro als auch in vivo hohe Aktivität gegen M. tuberculosis und ist somit ein vielversprechender Wirkstoffkandidat. Um einen tieferen Einblick in die Struktur-Aktivitäts-Beziehung zu erhalten und die Anwendbarkeit der RDC-Methode von kleinen Molekülen auf kleine und mittlere Peptide zu erforschen, wurde MGM mit NMR-spektroskopischen Mitteln untersucht. Bei der Analyse wurden NOE- und RDC (dipolare Restkopplungen)-restraints verwendet. Für die RDC-Analyse wurden anisotrope Proben in einem chemisch quervernetzten PDMS-Gel in CDCl3 angesetzt. In einer anschließenden simulated-annealing Berechnung mit XPLOR-NIH konnten niederenergetische Strukturen mit den ermittelten restraints berechnet werden. In einem Verfeinerungsprozess konnten zwei Strukturmodelle ermittelt werden, welche alle experimentellen Daten erfüllen. Mit den gleichen NMR-spektroskopischen Methoden wurde ein Fotoschalter (Dithienylcyclopenten-Derivat) und ein antibiotisches Cyclopentenonderivat (4,6-Diacetylhygrophorone A12) untersucht.

To get a deeper insight into the structure-activity relationship we used nuclear-Overhauser-effect- (NOE) and residual-dipolar-couplings- (RDC) restraints to determine the conformation of the MGM macrocycle in CDCl3. For the RDC-analysis we prepared anisotropic NMR-samples in a chemically cross-linked PDMS-gel. In a subsequent simulated-annealing calculation with XPLOR-NIH we used both NOE- and RDC-data as restraints. We could identify a structural bundle of low-energy structures which were in good agreement with NOE- and backbone RDC-restraints. After a structure refinement process by additionally using the proline side-chain RDCs we could identify two out of the 20 low-energy structures which are in good agreement with all experimental data. Same kind of NMR-measurement techniques were used to investigates a photoswitchable Dithienylcyclopentene derivative and the antibiotic cyclopentenone 4,6-Diacetylhygrophorone A12.
