Abstract:

Die enzymkatalysierte Verknüpfung von C–C-Bindungen mithilfe von Aldolasen und verwandten Enzymen ist aufgrund ihrer Enantio- und Diastereoselektivität von großem Interesse. Die Voraussetzung für eine umfangreiche Schutzgruppen-Chemie entfällt. Typischerweise sind diese Enzyme sehr flexibel und können eine breite Palette von elektrophilen Substraten akzeptieren. Hingegen haben sie eine sehr strenge Donor-Spezifität, welche ihre synthetischen Anwendungen begrenzt. Die Modifikation von Transaldolase B (TalBF178Y) und Fructose-6-phosphat-Aldolase (FSA) aus E. coli durch Protein-Engineering soll den Donorsubstrat-Anwendungsbereich erweitern. Unabhängig von der Substratstruktur katalysieren beide Enzyme die stereospezifische Carboligation in der D-threo Konfiguration. Es konnte eine Gruppe von verschiedenen 1-Hydroxy-2-alkanonen und DHA-Ether Derivaten als potentielle Donor-Substrate synthetisiert werden. Deren Reaktivität wurde für eine ausgewählte Anzahl von strukturbasiert ausgelegten FSA- und TalBF178Y-Varianten getestet. Durch die Verwendung von 3-Hydroxypropanal als Akzeptor konnte die Stereoselektivität der stabilen cyclischen hemiketalen Form leicht NMR-analytisch bestimmt werden. Die Untersuchungen der FSA- und TalBF178Y-Mutanten, die Beziehung zwischen Enzymaktivität und strukturellen Elementen der neuen Donorverbindungen sowie die Synthese neuartiger Desoxyzucker werden in dieser Arbeit diskutiert.

Enzymatic C–C bond forming reactions are highly attractive because of the unparalleled enantio- and diastereoselectivity of aldolases and related carboligating enzymes, without a requirement for extensive protective group chemistry. Typically, these enzymes are quite flexible to accept a structurally broad range of electrophilic substrates but have a rather strict specificity for their nucleophilic aldol donor substrate, which limits their range of synthetic applications. Thus, current persuasions to modify the donor substrate scope of transaldolase B (TalBF178Y) and fructose-6-phosphate aldolase (FSA) from E. coli by protein engineering are desirable. Independent of substrate structure, both enzymes catalyse a stereospecific carboligation resulting in the D-threo configuration. Synthesis of a panel of various 1-hydroxy-2-alkanone and DHA ether derivatives as potential donor substrates and investigation of their reactivity for a selected number of structure-based designed FSA variants were conducted. Using 3-hydroxy-propanal as the acceptor, the stereoselectivity was easily determined by NMR analysis of the stable cyclic hemiketal product isomers. The screening of the FSA mutants, the relationship between enzyme activity and the structural elements of the new donors, as well as the synthesis of new attainable classes of deoxysugars will be discussed in this thesis.
