Abstract:

In dieser Arbeit wurden die Promotoren der Gene p-gvpA, c-gvpA und fdx aus Halobacterium salinarum und des mc-gvpA-Gens aus Haloferax mediterranei mit dem bgaH-Reportergensystem untersucht. Die gvpA-Promotoren stammen aus den entsprechenden vac-Regionen, die für die Bildung von Gasvesikeln notwendig sind. Die Stärke der gvpA-Promotoren aus den drei vac-Regionen wurde quantifiziert und die Regulation ihrer Transkription näher untersucht. Dazu wurden die Promotoren mit dem bgaH-Leserahmen fusioniert und die basalen Aktivitäten der gvpA-Promotoren in Haloferax volcanii-Transformanten ermittelt. Es zeigte sich, dass der Promotor von p-gvpA (pA-Promotor) konstitutiv aktiv war, der Promotor von mc-gvpA (mcA-Promotor) eine geringe Aktivität besaß und der Promotor von c-gvpA (cA-Promotor) keine basale Aktivität hatte. Ebenso wurde die Aktivität der drei Promotoren bei Anwesenheit des GvpE-Aktivators in Hf. volcanii-Transformanten bestimmt. Es wurde dabei die Wirkung aller drei GvpE-Aktivatoren auf alle drei gvpA-Promotoren untersucht. Sowohl der mcA- als auch der pA-Promotor waren durch die drei GvpE-Proteine pGvpE, mcGvpE und cGvpE aktivierbar. Der cA-Promotor war dagegen nur durch das homologe cGvpE aktivierbar. Im Vergleich der Promotoren war der mcA-Promotor am stärksten aktivierbar, wohingegen der cA-Promotor am schwächsten aktivierbar war. An den A-Promotoren wurden auch die konservierten Sequenzen des BRE-Elements und der TATA-Box verändert, um zu testen ob diese Veränderungen einen Einfluss auf die basale Aktivität der Promotoren haben. Dem cA-Promotor, der eine kaum konservierte TATA-Box und ein kaum sichtbares BRE-Element besitzt, wurde durch Substitutionsmutation die gut konservierte TATA-Box, bzw. das gut konservierte BRE-Element des pA-Promotors, bzw. beide Elemente eingesetzt. Dieser mutierte cA-Promotor hatte zwar jeweils keine basale Aktivität, war aber besser durch GvpE aktivierbar. Somit genügten weder eine gut konservierte TATA-Box, noch ein gut konserviertes BRE-Element, noch die Kombination aus beiden, damit der cA-Promotor eine basale Aktivität zeigt. Alle Varianten führten aber zu einer verbesserten Aktivierbarkeit durch GvpE. Mit Hilfe von Mutationen wurde ein Bereich auf den gvpA-Promotoren identifiziert, der wichtig für die Aktivierung durch das GvpE-Protein ist. Es wurde ein chimärer pA-cA-Promotor, der die Sequenz des cA-Promotors, aber ein 21 nt langer DNA-Abschnitt strangauf der TATA-Box des cA-Promotors durch die entsprechende DNA-Sequenz des pA-Promotors ersetzt enthielt, konstruiert. In diesem 21 nt langen DNA-Abschnitt war auch das BRE-Element enthalten. Mit diesem pA-cA-Promotor konnte gezeigt werden, dass diese 21 nt lange pA-Promotor-Sequenz ausreichend für eine Aktivierung durch GvpE war und damit auch die GvpE Bindestelle beinhaltete. Es konnte ebenfalls die Wichtigkeit eines gut konservierten BRE-Elements bei der Aktivierung durch GvpE verdeutlicht werden. Der cA-Promotor enthält eine palindromische Sequenz strangauf der TATA-Box, die mutiert wurde, um den Wechselwirkungsbereich zwischen GvpE und Promotor näher zu untersuchen. Eine Deletion bewirkte den Verlust der Aktivierbarkeit des cA-Promotors. Substitutionsmutationen zeigten, dass bestimmte Nukleotide des Palindroms scheinbar unnötig für die Aktivierbarkeit waren, andere beeinflussten die Aktivierbarkeit. Der mcA-Promotor wurde ebenfalls durch Mutagenese analysiert. Der mcA-Promotor wurde mit Hilfe eines 7 nt deletionscanning über eine 49 nt lange Sequenz strangauf der TATA-Box analysiert. Es wurden 28 nt stromauf der TATA-Box, die auch das BRE-Element enthielten, identifiziert, die besonders wichtig für die Aktivierung durch GvpE waren. Auch ein Abtasten des Bereichs durch 4 nt lange Substitutionsmutationen über 34 nt strangauf des BRE-Elements konnte zeigen, dass ca. 20 nt angrenzend an das BRE-Element wichtig für die GvpE-Aktivierung waren. Damit wurden die Ergebnisse aus der Deletionsanalyse bestätigt. Auch der Einfluss der Salzkonzentration auf den mcA-Promotor aus Hf. mediterranei sowie auf den pA-Promotor und den fdx-Promotor aus Hb. salinarum wurde analysiert. Es stellte sich heraus, dass sowohl die basale Aktivität, als auch die durch mcGvpE induzierte Aktivität des mcA-Promotors bei 25% Salz im Medium höher war als bei 20% Salz. Die basale Aktivität des pA-Promotors und die durch pGvpE induzierte waren dagegen bei 20 und 25% Salz in etwa gleich. Bei 15% Salz war weder beim mcA- noch beim pA-Promotor Aktivität vorhanden. Der fdx-Promotor dagegen war bei allen drei Salzkonzentration aktiv.

In this thesis the promoters of the genes p-gvpA, c-gvpA and fdx of Halobacterium salinarum and of the mc-gvpA gene of Haloferax mediterranei were investigated with the bgaH reporter gene. The gvpA promoters were derived from the respective vac regions that are responsible for the formation of the gas vesicles. The strength of the gvpA promoters of the three vac regions were quantified and the regulation of their transcription was investigated. The promoters were fused with the bgaH reading frame to investigate the basal activities of the gvpA promoters using transformants of Haloferax volcanii. The promoter of p-gvpA (pA promoter) was highly active, the promoter of mc-gvpA (mcA promoter) only weakly active and the promoter of c-gvpA (cA promoter) was not active at all. Furthermore, the activities of the three gvpA promoters induced by all three GvpE activators were analysed. Both the mcA promoter and the pA promoter were activated by all three GvpE proteins, pGvpE, mcGvpE and cGvpE. In contrast, the cA promoter was only activated by the homologous cGvpE. The mcA promoter was most highly activated compared to the cA promoter which was the most weakly activated. The basal actvities of the gvpA promoters were compared to the ferredoxin promoter (fdx promoter) of Hb. salinarum. The fdx promoter is highly active, especially during exponential growth. The activity of the fdx promoter was a multiple times higher than the basal activity and the induced activity of the gvpA promoters. The two conserved sequences, the TATA box and the BRE element, of the the gvpA promoters were mutated to investigate the influence of these changes on the basal activities of the promoters. The TATA box and the BRE element of the cA promoter are only marginally conserved. If the TATA box or the BRE element of the cA promoter was replaced by the respective well conserved sequence of the pA promoter, or if the BRE element and the TATA box were exchanged for the respective sequences of the pA promoter, no basal activity of the cA promoter was detectable. In all cases, however, the cA promoter could be betteractivated by the GvpE protein. Therefore, neither a well conserved TATA box nor a well conserved BRE element nor a combination of the two were sufficient for mediating a basal activity. Nonetheless, all variations led to better activation by GvpE. Mutation analysis of the gvpA promoters showed a sequence important for the activation of the promoters by the GvpE protein. A chimeric pA-cA promoter was constructed with the cA promoter sequence in which the 21 nt upstream of the TATA box had been exchanged for the respective sequence of the pA promoter. The BRE element of the pA promoter was part of this sequence. The pA-cA promoter showed that the 21 nt of the pA promoter were sufficient for activation by GvpE and that a well conserved BRE element improved this activation. The cA promoter contains a further interesting element, a palindromic sequence upstream of the TATA box. This palindromic sequence was mutated for investigation of the interaction between the promoter and the GvpE activator. A deletion between the two palintromic parts led to a loss of the activation. Substitution mutagenesis of the palindromic sequence identified both significant and insignificant nucleotides for activation. The mcA promoter was also analysed by mutagenesis. The mcA promoter was analysed by a 7 nt deletion scanning extending over a 49 nt long sequence upstream of the TATA box. It turned out that the 28 nt upstream of the TATA box and including the BRE element were important for promoter activation. A 4 nt substitution walking extending over a 34 nt long sequence upstream of the BRE element identified a 20 nt sequence important for the activation of the promoter. Thus deletion scanning as well as substitution walking led to similar results. Furthermore, the influence of the salt concentration on the mcA promoter of Hf. mediterranei as well as the pA promoter and the fdx promoter of Hb. salinarum was investigated. It turned out that in the 25% salt medium both the basal activity and the activity induced by mcGvpE were higher than in 20% salt. In contrast, the basal activity as well as the activity induced by pGvpE of the pA promoter were nearly similar in the 20% and 25% salt. There was no activity of the mcA promoter and of the pA promoter detectable in the 15% salt medium. However, the fdx promoter was active in all three salt concentrations.
