Abstract:

Im Rahmen der vorliegenden Arbeit wurden 11 Bakterienstämme auf die Fähigkeit zur Bildung eines Prothrombin-Aktivators hin untersucht. Diese Untersuchung führte zur Entdeckung und Isolierung einer Prothrombin aktivierenden Aktivität aus dem fakultativ humanpathogenen Wasserkeim Aeromonas hydrophila, der Fischinfektionen und Durchfall beim Menschen verursacht. Datenbankrecherchen ergaben eine 90 %ige Sequenzidentität sowohl mit der Pro-Aminopeptidase Processing Protease aus Aeromonas caviae, als auch mit einer Elastase aus Aeromonas hydrophila. Der Prothrombin-Aktivator wurde biochemisch charakterisiert. Es handelt sich um eine einkettige Metalloprotease, die ein Molekulargewicht von 35 kDa und einen isoelektrischen Punkt bei pI = 4,4 aufweist. Die enzymatische Aktivität ist thermostabil und bleibt bis zu einer Temperatur von 55°C vollständig erhalten. Maximale Aktivität zeigt sich bei pH 6 in Gegenwart von 0 bzw. 1,5 M NaCl. Die Michaelis-Menten-Konstante KM und die maximale Reaktionsgeschwindigkeit vmax für die Prothrombin-Aktivierung betragen 1,77 (1,47) µM und 0,81 (1,66) mol/(min * mol Enzym) bei einer NaCl-Konzentration von 0 (1,5) M. Eine Analyse der Prothrombin-Spaltprodukte zeigte, dass unter anderem authentisches Thrombin entsteht, das seinerseits weiter abgebaut wird. Neben Prothrombin werden weitere Plasmaproteine (u.a. Fibrinogen und alpha2-Makroglobulin) gespalten. Ein fluorogenes Testsystem wurde etabliert, das die Bestimmung des Prothrombin-Gehalts in Plasma mit Hilfe der bakteriellen Protease mit hoher Sensitivität und Genauigkeit ermöglicht. Die vorliegenden Erkenntnisse lassen einen Einsatz des Enzyms als analytisches Werkzeug im Bereich der Gerinnungsdiagnostik als möglich erscheinen. Eventuell wäre es eine billigere und in größeren Mengen erzeugbare Alternative zu herkömmlichen Prothrombin-Aktivatoren aus Schlangengiften, die im Bereich der Gerinnungsanalytik eingesetzt werden, wie beispielsweise dem Ecarin aus dem Gift der Schlange Echis carinatus.

11 bacterial strains were analysed for their ability to produce a prothrombin activator. These investigations led to the identification and isolation of a prothrombin activator from the humanpathogenic germ Aeromonas hydrophila, which causes fish infections and diarrhoea in humans. The protein exhibits a sequence identity of 90% with both the pro-aminopeptidase processing protease from Aeromonas caviae and the so-called elastase from Aeromonas hydrophila. The prothrombin activator was characterized biochemically. It is a metalloprotease with a molecular weight of 35 kDa and an isoelectric point of pI=4,4. The enzymatic activity is fully retained up to a temperature of 55°C. The enzyme shows maximum activity at pH 6 and a NaCl-concentration of either 0 or 1,5 M. The Michaelis-Menten constant KM and the maximum velocity vmax for the activation of prothrombin are 1,77 (1,47) µM and 0,81 (1,66) mol/(min * mol enzyme) at a NaCl-concentration of 0 (1,5) M. An analysis of the degradation products of prothrombin revealed that authentic thrombin is generated, which is further processed. Besides prothrombin other plasma proteins (for instance fibrinogen and alpha2-macroglobuline) are also cleaved by the bacterial protease. A fluorogenic assay was established which allows the determination of the prothrombin concentration in plasma with the help of the bacterial enzyme with high sensitivity and accuracy. The enzyme seems to be a suitable for the usage as a diagnostic tool in the analysis of blood and blood products. It could possibly be a lower-priced alternative to prothrombin activators from snake venoms like ecarin from the Echis carinatus venom, which is commonly used in the analysis of coagulation.
