Abstract:

Das zunehmende Wissen über Zelloberflächen-Erkennungsmechanismen, wo Sialinsäuren eine essentielle Rolle spielen, bedingt einen steigenden Bedarf an Sialokonjugaten für biologische Studien. Um die entsprechenden Vorstufen in ausreichender Menge zur Verfügung zu stellen, stehen komplementäre chemische und enzymatische Methoden zur Verfügung. Die für die Synthese von Sialinsäuren bewährte Indium-vermittelte Allylierung von Aldosen nach der Barbier-Methode konnte erstmals auf kommerziell erhältliche Disaccharide mit gluco-Konfiguration am reduzierenden Ende zur Synthese von neuen 2-Desoxy-Zuckern bzw. 4,5-bisepi-KDN-Derivate übertragen werden. Dem gegenüber stehen die hoch interessanten enzymatischen Methoden, da diese bei breiter Substrattoleranz einen schnellen und bequemen Zugang zu einer Vielzahl modifizierter Sialinsäuren verschaffen. Mit dem rekombinantem E. coli-Stamm JM105/pKKNeuAc_3 konnte die Lyase des humanpathogenen Bakteriums Neisseria meningitidis neu produziert und für die präparative Synthese eingesetzt werden. Der eigentliche Aufbau zum Sialokonjugat erfordert zunächst die enzymatische Aktivierung der Sialinsäure mit anschließender Übertragung auf einen Akzeptor. Mit dem rekombinanten E. coli-Stamm JM105/pKKCMP konnte die neu produzierte CMP-Sialat-Synthetase aus Neisseria meningitidis zur Synthese weiterer CMP-Konjugate, die an C-5-Position modifizierter Sialinstrukturen enzymatisch im präparativen Maßstab zu den entsprechenden erfolgreich genutzt werden. Mit einem weiteren Enzym, der α-2,3-Sialyltransferase aus Campylobacter jejuni, gelang es, die aktivierten CMP-Konjugate auf fluoreszenzmarkierte Akzeptorsubstrate zu übertragen. Als Fluoreszensmarker wurde ein Acridon-Anker eingesetzt, der sich als besonders stabil erwies und hoch empfindlich detektierbar ist. Die Akzeptorsubstrate wurden hierbei nach chemischen oder chemoenzymatischen Methoden hergestellt und auf ihre Verwendbarkeit in enzymatischen Reaktionen hin getestet.

The increasing knowledge about the mechanisms of cell surface recognition, where sialic acids play an essential role, stimulates an increasing requirement of sialic acid conjugates for biological studies. Several complementary chemical and enzymatic methods have been developed to provide the appropriate precursors in adequate amount. The established indium-mediated allylation of aldoses according to the Barbier-method could be transferred for the first time to commercially available disaccharides with gluco configuration at the reducing end, to allow for the synthesis of new 2-deoxy sugars and 4,5-bisepi-KDN-derivatives, respectively. On the other hand, enzymatic methods are high of interest, because this leads to a diversity multiplicity of modificated sialic acids in a short and convenient way when using enzymes that have a broad substrate tolerance. From recombinant E. coli strain JM105/pKKNeuAc_3 the lyase of the human pathogenic bacteria Neisseria meningitidis could be produced and utilized for preparative syntheses. The actual build-up of a sialoconjugate first demands the enzymatic activation of the sialic acid, followed by its transfer to an acceptor. From recombinant E. coli strain JM105/pKKCMP the newly produced CMP-sialate synthetase from Neisseria meningitidis could be successfully applied to the further enzymatic synthesis of CMP-conjugates at a preparative scale using C-5 modified sialic acids. By using the α-2,3-sialyltransferase from Campylobacter jejuni as a further enzyme, the activated CMP-conjugates could be transferred to fluorescence labeled acceptor substrates. An acridon anchor was used as fluorescence marker, which proved to be stable and highly sensitive for analytical proposes. The acceptor substrates were synthesized by chemical or chemoenzymatic methods and also tested for their practicability in enzymatic reactions.
