Abstract:

Ein vielversprechender Ansatz zur Synthese von Peptidmimetika ist das Konzept der Präsentation pharmakophorer Gruppen am Ketidrückgrat. Dafür ist eine iterative asymmetrische Aldoladdition am polymeren Träger zur kombinatorischen Synthese der Ketide gewünscht. Durch geeignete Reaktionsbedingungen unter Verwendung chiraler Auxiliare ist die Kontrolle sowohl der relativen als auch der absoluten Konfiguration an den bei der Reaktion gebildeten stereogenen Zentren möglich. Die Verwendung chiraler 1,2-N-O-Heterocyclen als Auxiliare in der Aldoladdition eröffnet die Möglichkeit der Erzeugung des für die nächste Aldoladdition notwendigen Aldehyds in einem Schritt. In dieser Arbeit wurden die Synthesen neuer mono- und bicyclischer Isoxazolidine und Oxazinane als Vertreter dieser Stoffklasse entwickelt und durchgeführt. Die Eignung der dadurch erhaltenen 1,2-N-O-Heterocyclen als Auxiliare in der asymmetrischen Aldoladdition wurde anhand der Reaktionen ihrer auf Bor, Lithium und Titan basierenden Enolate untersucht. Durch diese Untersuchungen wurde einerseits das Wissen um die Synthesemöglichkeiten für chirale 1,2-N-O-Heterocyclen erweitert; andererseits wurden strukturbasierte Beschränkungen sowie erzielbare simple und induzierte Diastereoselektivitäten bei der in der Literatur kaum bekannten Aldoladdition von Vertretern dieser Substanzklasse ermittelt.

A promising strategy for the design of peptido-mimetics is to present pharmacophoric moieties attached to a poly ketide backbone. The combinatorial synthesis of the poly ketides can be achieved by an iterative asymmetric aldol addition on a polymeric support. By adjusting the reaction parameters and utilisation of chiral auxiliaries the relative as well as absolute configuration of the new stereogenic centres can be controlled. If 1,2-N-O-heterocycles are used as auxiliaries in the aldol addition, the aldehyde function, which is necessary to enter the next reaction cycle, can be (re-)generated in one single step. In the context of this work the syntheses of novel mono- and bicyclic isoxazolidines as well as oxazinanes, being representatives of the aforementioned structural class, have been elaborated and optimized. The suitability of the generated auxiliaries in asymmetric aldol additions was tested by reacting the respective Boron-, Lithium- and Titanium-enolates with benzaldehyde. These investigations substantially increased the knowledge on synthesis routes yielding 1,2-N-O-heterocycles. In addition, the scope and scaffold related limitations were elucidated as well as the simple and induced diastereoselectivity in aldol additions of this rarely known substance class.
