Abstract:

In dieser Arbeit wurde gezeigt, dass die Multifrequenz Elektronen Paramagnetische Resonanz (EPR), Elektron-Kern-Doppelresonanz (ENDOR) und HYSCORE (hyperfine sublevel correlation) Spektroskopie für eine Strukturaufklärung des biologisch relevanten Kupfer-Ions und Chelidamat-Liganden sehr nützlich ist. Die Kombination der Multifrequenz EPR-Methoden mit computer-gestützten numerischen Simulationen ermöglichten eine vollständige Aufklärung der Struktur des Cu(II)-Chelidamat Komplexes. Die strukturelle Charakterisierung des Cu (II)-Chelidamat Komplexes wurde mittels Hochfeld-EPR, klassischer cw (continuous wave) EPR und zwei-dimensionaler EPR durchgeführt. Die Orientierung der g- Matrix innerhalb des Moleküls und seine Hauptachsenwerte wurden bestimmt. Im Vergleich zum konventionellen X-Band (9.7 GHz) erlaubt die Hochfeld-EPR bei 180 GHz (G-Band) eine genauere Bestimmung von den g-Matrix-Elementen. Ausserdem wurden in dieser Arbeit Kupfer und Stickstoff Hyperfein-Wechselwirkungen durch Multifrequenz cw EPR und Puls Elektronspin-echo-Experimente untersucht. Diese Experimente ermöglichten einen Einblick in die Koordinationsgeometrie der Komplexierung des Cu-Ions mit Chelidamsäure als Liganden. Eine genaue Untersuchung der Komplexierung des Kupfer(II)-Zentrums und seinen koordinierten Liganden wurde mit Puls ENDOR, ESEEM (electron spin echo envelope modulation) und HYSCORE Methoden durchgeführt. Um zuverlässige und genaue Informationen über die Hyperfein- und Kernquadrupol-Wechselwirkungen sowohl stark als auch schwach gekoppelter Kerne zu erhalten, wurden orientierungs-selektive hochauflösende ENDOR- und HYSCORE-Experimente durchgeführt. Die Puls-ENDOR und HYSCORE Spektroskopie von Isotopen-markierten 14N- und 15N-Kernen ermöglichte die Identifizierung der koordinierten Liganden. Durch orientierungs-selektive HYSCORE Messungen wurden der Hyperfein- und Kernquadrupol-Tensor vollständig bestimmt. Darüber hinaus konnte gezeigt werden, dass das Lösungsmittel Dimethylformamid (DMF) über den Sauerstoff koordiniert wird. Die 1H-Hyperfein-Wechselwirkung des Chelidamat-Liganden wurde mit Multifrequenz Puls ENDOR Methoden untersucht. Im Hochfeld (W-Band) wurden in 1H-ENDOR Spektren Polarisationseffekte beobachtet. Diese Informationen, die nur mit der Hochfeld-ENDOR Spektroskopie erhältlich sind, wurden verwendet um das Vorzeichen der Hyperfein-Kopplung zu bestimmen. Die Daten von EPR, ENDOR und HYSCORE Messungen wurden mit der Geometrie-optimierten Struktur des Cu(II)-Chelidamat Komplexes aus Dichte-Funktional-Theorie (DFT) Berechnungen verglichen. Die abgeleitete Struktur bei 10 K liegt auch bei Raumtemperatur in der flüssigen Lösung vor, wie aus einer Bestimmung seines effektiven Volumens abgeleitet wurde.

In this thesis it is demonstrated that multifrequency electron paramagnetic resonance (EPR), electron nuclear double resonance (ENDOR) and hyperfine sublevel correlation (HYSCORE) spectroscopy are very useful for a structure investigation of the biologically relevant copper ion and chelidamate ligand. The combination of multifrequency EPR methods with computer-supported numerical simulations allowed for an entire elucidation of the structure of the Cu(II)-chelidamate complex. Structural characterization of a Cu(II)-chelidamate complex was performed by high-field EPR, classical continuous wave (cw) EPR and two dimensional EPR. The orientation of the g-matrix within the molecule and its principal axies values were determined. In comparison to conventional X-band (9.7 GHz) the high-field EPR with 180 GHz (G-band) allowed a more accurate determination of the g-matrix elements. Moreover, in this study copper and nitrogen hyperfine interactions were investigated by multifrequency cw EPR and pulse electron spin echo experiments. These experiments allowed for an insight into the coordination geometry of copper complexation with chelidamic acid as a ligand. An accurate investigation of the complexation for the copper(II) and its co-ordinated ligands was studied with pulse ENDOR, electron spin echo envelope modulation (ESEEM) and HYSCORE methods. To obtain reliable and precise information about the hyperfine and nuclear quadrupole interactions of both strongly and weakly coupled nuclei, orientation-selective high-resolution ENDOR and HYSCORE experiments were performed. The pulse ENDOR and HYSCORE spectroscopy of isotopic labeled 14N and 15N nuclei allowed for an identification of the co-ordinated ligands. By orientation-selective HYSCORE measurements hyperfine and nuclear quadrupole tensors were completely determined. In addition, it could be shown that the solvent dimethylformamide (DMF) is co-ordinated to the copper ion via the oxygen. The 1H hyperfine interaction of the chelidamate ligand was investigated by multifrequency pulse ENDOR methods. In the high-field W-band 1H-ENDOR spectra polarization effects were observed. This information, only obtainable by high-field ENDOR spectroscopy was used to determine the sign of the hyperfine coupling. The data from EPR, ENDOR and HYSCORE measurements were compared to the geometry-optimized structure of the Cu(II)-chelidamate complex revealed by density functional theory (DFT) calculations. The structure derived at 10 K apparently also persists at room temperature in liquid solution, as was deduced from a determination of its effective volume.
