Abstract:

Depressive Erkrankungen werden, laut einer WHO-Studie, bis zum Jahr 2020 den zweiten Platz unter den Krankheiten, welche die Lebensqualität beeinflussen einnehmen. Das von Merck KGaA, Darmstadt als Antidepressivum entwickelte Vilazodone besitzt einen dualen Wirkmechanismus und wird gegenwärtig von dem amerikanischen Pharmacogenomics-Unternehmen Genaissance in die klinische Phase III geführt. Der SSRI und 5-HT1A-Agonist hat mit seinen subnanomolaren IC50-Konzentrationen die Voraussetzungen ein erfolgreicher Wirkstoff gegen Depressionen zu werden. Für eine erfolgreiche Zulassung als Medikament ist es unumgänglich auch die Metaboliten von Vilazodone pharmakologisch und toxikologisch zu untersuchen. Bis zum Beginn der vorliegenden Dissertation war die Synthese der beiden Hauptmetaboliten nicht gelungen. Ziel der Arbeit war es, die für den Aufbau der Metabolite von Vilazodone benötigten neuartigen C3-funktionalisierten 6-Hydroxy- sowie 4-Hydroxyindol-5-carbonitrile zu synthetisieren, um anschließend die beiden Hauptmetaboliten bereitstellen zu können. Zusätzlich sollte eine Synthese zu den bisher nicht zugänglichen 6-Hydroxy- und 4-Hydroxyindol-5-carbonsäuren mit C3-Funktionalisierung etabliert werden. Von diesen Indolen mit Salicyl-Charakter erhofft man sich eine bessere Löslichkeit unter physiologischen Bedingungen. Darüber hinaus können die Ester und Ether dieser Indolderivate interessante Prodrug-Eigenschaften haben. Im ersten Teil der Dissertation wird zunächst das serotonerge System beschrieben und die Wirkmechanismen der gängigen Wirkstoffklassen erläutert. Anschließend werden einige Vertreter dieser Wirkstoffklassen beschrieben bevor Vilazodone und seine Metaboliten vorgestellt werden. Im zweiten Teil werden zunächst denkbare Synthesen zu den geforderten Indolen beschrieben und dann eine geeignete Synthesestrategie entworfen. Anhand dieser Strategie wird anschließend die Synthese der 6-Hydroxy- und 4-Hydroxyindol-5-carbonitrile, der 6-Hydroxy- und 4-Hydroxyindol-5-carbonsäuren sowie der Metaboliten von Vilazodone und weiterer interessanter Indolderivate mit den jeweiligen Substitutionsmustern beschrieben. Darüber hinaus werden kurz einige biologische Daten sowie gemessene und berechnete Löslichkeiten diskutiert. Die Arbeit schließt mit einem detaillierten experimentellen Teil und der Auswertung der angefertigten Spektren zum Strukturbeweis.

Depressive disorders are expected to rank on the second position of the greatest causes of diseases in the year 2020 (DALY study, WHO). The antidepressant Vilazodone, which was developed by Merck KGaA, Darmstadt has a dual operation mechanism and is currently lead to clinical phase III by the pharmacogenomics company Genaissance. With subnanomolares IC50-concentrations has the 5-HT1A-agonist and SSRI good conditions to get a successful antidepressant. For the admission as drug it is necessary to investigate pharmacological and toxicological characteristics of the metabolites, as well. Up to the start of this work the synthesis of the metabolites of Vilazodone was not a success. Target of this work was the synthesis of newly C3-functionalized 6-Hydroxy- and 4-Hydroxyindole-5-carbonitriles for the build up of the two main metabolites and the synthesis of these, as well. Additionally it should established an access to not known 6-Hydroxy- and 4-Hydroxyindole-5-carbonacids with C3-function. These Indoles with salicylic character could have a better solubility under physiological terms. Beyond this esters and ethers of this Indoles could have interesting produg conditions. In the first section of this work is described the serotoninergic system and the mechanism of action of common drug classes against depressive disorders. Adjacent are presented some common drugs of these classes and Vilazodone with its metabolites is introduced. In the second section are described possibilities for the build up of the needed Indoles and it is outlined a strategy of synthesis. With this strategy is then described the synthesis of 6-Hydroxy- and 4-Hydroxyindole-5-carbonitriles, of 6-Hydroxy- and 4-Hydroxyindolecarbonacids as well as the metabolites of Vilazodone and further interesting Indoles with this pattern of substitution. Beyond these are shortly discussed biological facts and solubilities. The work closed with a detailed experimental section and an evaluation of the spectroscopic facts as proof of structure.
