Abstract:

In dieser Dissertation konnte der Mechanismus der scherstressabhängigen Apoptoseinduktion bei vaskulären Endothelzellen (HUVEC) aufgeklärt werden. Unter laminarem Scherstress liegt der Rezeptorkomplex aus CD47 und Integrin alpha V beta 3 in nicht aktivierbarer Konformation vor. Nach Wegfall des laminaren Scherstresses kommt es zu einer Konformationsänderung von CD47, die eine Aktivierung des Rezeptorkomplexes und die Auslösung der Apoptose durch Bindung von Thrombospondin 1 ermöglicht und in Folge die Expression des Fas Liganden einleitet. Die Apoptose erfolgt über den Fas Signalweg. Die Konformationsänderung von CD47 wurde durch Untersuchung der scherstressabhängigen Transkriptions- und Translationsregulation von CD47 über quantitative PCR, Northern Blot, Western Blot, indirekte Immunfluoreszenzfärbung und Apoptoseinhibitionsstudien nachgewiesen. Die Beteiligung des Fas Signalwegs an der scherstressabhängigen Apoptose wurde durch Apoptoseinhibitionsstudien mit monoklonalen Antikörpern gegen Fas Ligand und Western Blot Untersuchungen belegt.

In this work the mechanism of shear stress dependent apoptosis of vascular endothelial cells (HUVEC) could be solved. At laminar shear stress the receptor complex of CD47 and Integrin alpha V beta 3 can not be activated by binding Thrombospondin 1. After cease of laminar shear stress CD47 unterlies a conformational change that allows Thrombospondin 1 to bind to the receptor complex and to induce apoptosis by expression of Fas ligand. Apoptosis results from Fas signaling pathway. The conformational change of CD47 was proven from shear stress dependent regulation of CD47 transcription and translation by quantitative PCR, Northern blot, Western blot, indirect immunefluorescence studies, and inhibition of apoptosis. The participation of the Fas signaling pathway in shear stress dependent apoptosis was verified by inhibition of apoptosis with monoclonal antibodies against Fas ligand and Western blot analysis.
