Abstract:

In der vorliegenden Dissertation wurden ATP-Synthasen, OxPhos-Komplexe und Superkomplexe in Mitochondrien von Säugetieren und der Grünalge Chlamydomonas reinhardtii funktionell analysiert und die Chloroplasten-ATP-Synthase aus Spinat (Spinacia oleracea) strukturell charakterisiert. Für die Untersuchung der Komplexe der oxidativen Phosphorylierung (OxPhos-Komplexe) von Säugetieren wurden Mitochondrien aus Herzgewebe von Rind, Schwein, Wildschwein, Reh, Maus, Ratte (Wistar und Fischer) und aus Nierengewebe von Reh isoliert. Die Enzymkomplexe wurden mit dem milden Detergens Digitonin aus der Lipidmembran gelöst (solubilisiert) und mittels einer blau-nativen Gelelektrophorese (BN-PAGE) nach ihrer Größe aufgetrennt. Der Vorteil dieser Methode liegt darin, dass die enzymatische Aktivität und die dreidimensionale Struktur der Proteine ebenso wie Protein-Protein Wechselwirkungen erhalten bleiben. Eine Analyse der Atmungskettenkomplexe erfolgte durch In-Gel Aktivitätstests. Bei diesen Tests katalysiert der enzymatisch aktive Proteinkomplex die Bildung eines farbigen Präzipitats, dass auf der jeweiligen Gelbande detektiert werden kann. Bei den OxPhos-Komplexen aus isolierten Rinderherzmitochondrien war wie in vorherigen Arbeiten von Schäfer et al. zu beobachten, dass sich die Komplexe I, III2 und IV zu übergeordneten Strukturen, den sogenannten Superkomplexen (SC), zusammenlagern. In der vorliegenden Arbeit konnte gezeigt werden, dass auch bei Mitochondrien aus Schwein-, Wildschwein-, Reh-, Mäuse-, Rattenherz und Rehniere die Superkomplexe III2IV1, I1IV2, I1III2 (a), I1III2IV1 (b), I1III2IV2 (c) und I1III2IV3 (d) auftreten. Anhand der Intensität der Coomassie-gefärbten Banden und des gebildeten Präzipitats war zu erkennen, dass Komplex I hauptsächlich in Form von Superkomplexen gebunden ist und Komplex IV zu einem großen Teil als individueller Komplex vorliegt. Aus diesen In-Gel Aktivitätstests wurden mit der Software Delta 2D die relativen spezifischen Aktivitäten der Komplexe I und IV in dem BN-Gel ermittelt. Bei allen untersuchten Säugetierproben besaßen die Superkomplexe c und d für Komplex I die höchste spezifische Aktivität. Der Vergleich zwischen der spezifischen Aktivität des individuellen Komplexes I und dieser Superkomplexe zeigte, dass bei Rind, Schwein, Wildschwein und Reh die Aktivitäten im SC um den Faktor 2,3 gesteigert waren. Bei Maus und Ratte führte eine Assemblierung zu Superkomplexen bei I1III2IV2 (c) zu einer Zunahme der Aktivität um den Faktor 3,4 und bei I1III2IV3 (d) um den Faktor 3,6. Ein deutlicher Anstieg der Aktivität im Vergleich zum individuellen Komplex I war für Superkomplex c (Faktor 12,5) und Superkomplex d (Faktor 11,6) aus Rehniere zu beobachten. Bei den Komplex IV-Aktivitätstests besaß bei allen Säugetierproben der individuelle Komplex IV die mit Abstand niedrigste Aktivität. Die Superkomplexe b und c zeigten bei Rind, Schwein, Wildschwein, Reh und Ratte die höchste spezifische Aktivität, bei Maus traf dies nur auf Superkomplex b zu. Diese deutliche Steigerung der Aktivität stellte sich im Vergleich zu dem individuellen Komplex IV wie folgt dar: Bei Herzmitochondrien aus Rind, Schwein, Wildschwein, Reh und Maus für Superkomplex b eine Zunahme um den Faktor 8,3 und für Superkomplex c um den Faktor 8,0 (ohne Maus). Bei den Rattenherzmitochondrien ist I1III2IV1 (b) um den Faktor 11,6 und I1III2IV2 (c) um den Faktor 14,02 aktiver. Bei den Solubilisaten aus Rehniere zeigte sich ebenfalls eine Steigerung der Aktivität für Superkomplex b (Faktor 4,2) und Superkomplex c (Faktor 4,8). Mit Hilfe von Antikörpern gegen die Komplexe I, III, IV und V konnten in 1-D Western Blots von blau-nativen Gelen sowohl die individuellen Komplexe als auch höhere Assemblierungen nachgewiesen werden. In Übereinstimmung mit den Aktivitätstests bestätigte sich, dass Komplex I enthaltende Proteinbanden im Bereich der Superkomplexe deutlich stärkere Signale lieferten als der individuelle Komplex. Bei den Komplexen III2 und IV war dies nicht zu beobachten, die Signale für die individuellen Komplexe waren sehr stark ausgeprägt. Für Komplex V, die ATP-Synthase, konnten sowohl das Monomer und Dimer als auch höhere Oligomere bei allen Proben gefunden werden. Die Solubilisate aus Mäuse-, Schwein-, Wildschwein-, Rehherz und Rehniere wurden mit Hilfe von 2-D BN/SDS-PAGE gelelektrophoretisch aufgetrennt und die mitochondrialen Proteine durch "Peptide Mass Fingerprint" (an einem MALDI-TOF MS) identifiziert. Da für Rinderherzmitochondrien schon entsprechende Experimente von Kratochwil und Hunzinger et al. bzw. für Rattenherzmitochondrien von Reifschneider et al. durchgeführt worden sind, wurde bei diesen Proben auf eine weitere Analyse verzichtet. Ebenso wurden von Groebe et al. die mitochondrialen Proteine von Mensch und Podospora anserina mittels MALDI-MS untersucht. In der vorliegenden Arbeit konnten abhängig vom Gewebe zwischen 29 und 36 mitochondriale Proteine identifiziert werden, hierbei handelte es sich bei ca. 80 % um Untereinheiten der Komplexe I, III, IV und V der oxidativen Phosphorylierung. Es war zu beobachten, dass ein Großteil der identifizierten Untereinheiten der OxPhos-Komplexe eine sehr ähnliche elektrophoretische Mobilität besitzen. Ebenso zeigte ein Vergleich mit den bei Rinder- und Rattenherz gefundenen Resultaten diese Übereinstimmungen. So konnten besonders bei der ATP-Synthase (Komplex V) der untersuchten Gewebe Untereinheiten für das Monomer, das Dimer und andere Oligomere gefunden werden. Mittels 2-D Western Blots wurden weitere Untereinheiten der OxPhos-Komplexe nachgewiesen. Für den Komplex II zeigte sich, dass dieser unter den gewählten Bedingungen (2-D BN/SDS-PAGE) nicht an der Bildung von Superkomplexen beteiligt ist und nur als individueller Komplex vorliegt. Für eine quantitative Analyse der identifizierten Proteinspots wurden 2-D Gele von den Säugetierproben mit dem Fluoreszenzfarbsoff Sypro® Ruby gefärbt. Die Auswertung der Gele erfolgte mit der Software Delta2D. Die Daten der Herzgewebe von Rind, Schwein, Wildschwein und Reh wurden miteinander verglichen. Die Gesamtproteinmenge für die Komplexe I, III und V lag bei diesen Proben in einem ähnlichen Bereich, es zeigten sich lediglich kleine Unterschiede bei den Verhältnissen zwischen dem individuellen Komplex und den Superkomplexen. Des Weiteren war zu beobachten, dass die ATP-Synthase in einem Bereich von 61 bis 74 %, abhängig vom Organismus, in monomerer Form vorliegt. Die größten Unterschiede waren bei der Gesamtproteinmenge von Komplex IV zu beobachten. Bei dem Vergleich zwischen Rinder-, Mäuse- und Rattenherz traten zwischen diesen unterschiedlichen Organismen teilweise deutliche Unterschiede für die OxPhos-Komplexe I und V auf. So konnte für Ratte nachgewiesen werden, dass im Gegensatz zu den anderen untersuchten Geweben die Proteinmengen für das Monomer V1 und das Dimer V2 der ATP-Synthase nahezu identisch sind. Bei Maus war zu erkennen, dass Komplex V zu mehr als 78 % in Form von V1 vorliegt. Ebenso war bei Maus zu beobachten, dass Komplex I hauptsächlich in Superkomplex a gebunden ist. Sowohl bei Rind und Ratte, als auch bei den zuvor untersuchten Geweben (Reh, Schwein, Wildschwein) war immer Superkomplex b favorisiert. Von den Dimeren und höheren Oligomeren von Komplex V ist bekannt, dass sie an der Gestaltung der Morphologie der Cristae beteiligt sind. Abgesehen davon steht bisher noch nicht fest, inwieweit die enzymatische Aktivität durch diese Oligomerisierung beeinflusst wird. Daher wurde für eine weitere Untersuchung der ATP-Synthase der In-Gel ATP-Hydrolyse-Aktivitätstest verwendet. Erste Versuche zeigten, dass die BN-PAGE ungeeignet war. Offensichtlich beeinträchtigte der Farbstoff Coomassie Brillant Blau die Zugänglichkeit des Substrats und auch die Detektion des gebildeten Präzipitats, daher wurde die mildere farblos-native Gelelektrophorese (CN-PAGE) für weitere Experimente ausgewählt. So konnte anhand dieser Aktivitätstests erstmals nachgewiesen werden, dass bis dato noch nicht untersuchte Säugetierproben (Reh-, Schwein-, Wildschweinherz) Oligomere der ATP-Synthase ausbilden, die eine Hydrolyse-Aktivität zeigen. Für die mitochondrialen Solubilisate aus Maus, Ratte und Rind wurden ähnliche Resultate gefunden. Im Gegensatz zu den in der Literatur gefundenen Ergebnissen konnte gezeigt werden, dass die monomere Form die höchste Hydrolyse-Aktivität besitzt und mit steigendem Assemblierungsgrad die Aktivität abnimmt. In welchem Zusammenhang die Hydrolyse mit der Synthese von ATP durch Komplex V steht ist bisher noch nicht vollständig aufgeklärt. Davon ausgehend, dass eine Verringerung der Hydrolyse-Aktivität mit einer Zunahme der ATP-Synthese korreliert, würde die Syntheserate mit steigendem Assemblierungsgrad deutlich zunehmen. Dementsprechend könnte dadurch gezielt die enzymatische Aktivität der ATP-Synthase innerhalb einer Zelle gesteuert werden ohne die de novo Expression dieses Enzymkomplexes zu verändern. Untersuchungen von Meyer zu Tittingdorf et al. und Hema et al. haben gezeigt, dass äußere Stressfaktoren den metabolischen Zustand der Mitochondrien der Grünalge Chlamydomonas reinhardtii beeinflussen. Im Rahmen dieser Arbeit sollte analysiert werden, welchen Einfluss unterschiedliche Temperaturen während der Wachstumsphase auf die Organisation und Funktion der OxPhos-Komplexe ausüben. Für die Kultivierung der Alge wurden zwei unterschiedliche Wachstumsbedingungen untersucht. Bei photomixotrophem Wachstum wird dem Kultivierungsmedium Acetat als Kohlenstoffquelle zugesetzt, bei photoautotrophem Wachstum muss Kohlenstoff aus der Luft, in Form von CO2, assimiliert werden. Für eine funktionelle Analyse der OxPhos-Komplexe I und IV wurden die oben beschriebenen In-Gel Aktivitätstests eingesetzt. Ähnlich wie auch bei den Mitochondrien in Säugetieren assemblieren die Komplexe I, III und IV in C. reinhardtii zu Superkomplexen, I1III2IV1 (S1) und I1III2IV2 (S2). Unter photomixotrophen Bedingungen zeigte sich, dass eine Erhöhung der Temperatur zu einer deutlichen Steigerung der Aktivität von Komplex I (1,6-fach) und IV (1,8-fach) in den Superkomplexen S1 und S2 führte. Für die individuellen Komplexe war dieser Effekt nicht zu beobachten. Ein ähnlicher Trend war unter photoautotrophem Wachstum für Komplex I zu erkennen. Demgegenüber führte bei Komplex IV eine erhöhte Temperatur zu einer kleinen Verringerung der spezifischen Aktivität der Superkomplexe. Durch einen Vergleich zwischen der Aktivität des individuellen Komplexes und der Superkomplexe S1 und S2 konnte erstmals auch für einen pflanzlichen Organismus gezeigt werden, dass eine Assemblierung zu Superkomplexen zu einer deutlichen Steigerung der enzymatischen Aktivität führt. Für Komplex I äußerte sich dies in einer 5-fachen und für Komplex IV in einer 10-fachen Zunahme der Aktivität. Die Ergebnisse der vorliegenden Dissertation können wie folgt zusammengefasst werden: Sowohl in Mitochondrien aus Herzgewebe von Säugetieren als auch in den Mitochondrien von C. reinhardtii konnte eine Assemblierung der Komplexe I, III und IV der oxidativen Phosphorylierung zu Superkomplexen nachgewiesen werden. Bei allen untersuchten Organismen war bei den Superkomplexen eine deutliche Zunahme der enzymatischen Aktivität im Vergleich zum individuellen Komplex zu beobachten. Für die ATP-Synthase (Komplex V) konnte bei den Säugetierproben gezeigt werden, dass über den Grad der Oligomerisierung die Hydrolyse-Aktivität beeinflusst werden kann. In Zusammenarbeit mit den Forschungsgruppen von Prof. Dr. Grüber (NTU, Singapur) und Prof. Dr. Gordeliy (IBS Grenoble; ICS Jülich; MIPT Moskau) sollte mittels Röntgenkristallographie die Struktur der Chloroplasten-ATP-Synthase aufgeklärt werden. Es gibt Kristallstrukturbilder mit hoher Auflösung von dem hydrophilen F1-Teil und von einigen isolierten Untereinheiten des membranintegralen FO-Teils, jedoch ist es bisher noch nicht gelungen den vollständigen und intakten Enzymkomplex zu kristallisieren. Zunächst wurde von mir die Chloroplasten-ATP-Synthase aus Spinat (Spinacia oleracea) in größeren Mengen (100-200 mg) isoliert. Mittels BN-PAGE konnte nachgewiesen werden, dass das Enzym intakt war. Die Synthese-Aktivität der isolierten ATP-Synthase wurde nach Einbau in Liposomen und Applikation eines elektrochemischen Protonengradienten (∆Ψ+∆pH) in einem anschließenden Luciferin-Luciferase-ATP-Test ermittelt. In der Arbeitsgruppe von Prof. Dr. Grüber wurde von Dr. Asha Manikkoth Balakrishna die Kristallisation mittels Dampfdiffusionsverfahren durchgeführt. Die erhaltenen Kristalle wurden an einem Synchrotron (Swiss Light Soure) am Paul-Scherrer-Institut (Schweiz) analysierst. Es zeigte sich, dass bis auf den c14-Zylinder der ATP-Synthase alle anderen Untereinheiten während des Kristallisationsprozesses verloren gingen, die ermittelte Kristallstruktur besaß eine Auflösung von 4,50 Å. In der Arbeitsgruppe von Prof. Dr. Gordeliy erfolgte die Kristallisation mittels kubischer Lipid-Phase. Obwohl auch hier von einer intakten ATP-Synthase ausgegangen worden ist, konnten unter diesen Bedingungen ebenfalls nur Kristalle des c14-Zylinders erhalten werden, allerdings erstmals mit einer Auflösung von 2,48 Å.

This thesis focuses on the functional analysis of ATP synthases, OxPhos complexes and supercomplexes in mitochondria of mammals and of the green algae Chlamydomonas reinhardtii and the structural characterization of the chloroplastidic ATP synthase from spinach (Spinacia oleracea). For the analysis of mammalian OxPhos complexes mitochondria from bovine, pig, wild boar, deer, mouse and rat (Wistar and Fischer) heart and deer kidney were isolated. The enzyme complexes were detached from the lipid bilayer with the mild detergent digitonin (solubilized) and separated by blue-native electrophoresis (BN-PAGE). The advantages of this method are the preservation of the enzymatic activity, of the three-dimensional native structure and of the protein-protein interactions. The characterization of OxPhos complexes was performed by in-gel activity tests for complexes I and IV. In these tests the enzymatic active protein complex catalyses the formation of a coloured precipitate that can be detected for every band in the gel. Previous studies of Schäfer et al. on bovine heart mitochondria demonstrated that the OxPhos complexes I, III2 and IV are assembling to higher-ordered structures, so called supercomplexes (SC). In this thesis the supercomplexes III2IV1, I1IV2, I1III2 (a), I1III2IV1 (b), I1III2IV2 (c) und I1III2IV3 (d) were detected in mitochondria from bovine, pig, wild boar, deer, mouse, rat heart and deer kidney. The Coomassie staining of the BN-gels and the activity tests indicated that complex I is mainly associated in supercomplexes while complex IV mostly exists as individual complex. For quantitative analysis the relative specific activity of complex I and IV containing protein bands were determined by the software Delta2D. In all examined mammalian tissues the supercomplexes c and d (for complex I) possessed by far the highest specific activity. Comparison of the specific activity of the individual complex I and the supercomplexes revealed that the activity in I1III2IV2 (c) and I1III2IV3 (d) in bovine, pig, wild boar and deer was increased 2.3-fold. In the mouse and rat mitochondria the assembling of supercomplexes showed an increase in activity for I1III2IV2 3.4-fold and for I1III2IV3 3.6-fold. A significant enhancement of activity in comparison with the individual complex was observed in supercomplex c (12.5-fold) and supercomplex d (11,6-fold) from deer kidney. The in-gel activity test for complex IV demonstrated that individual complex IV exhibited by far the lowest specific activity in all examined mammalian samples. The supercomplexes b and c showed for bovine, pig, wild board and deer the highest specific activity, for the mouse samples this was only observed in supercomplex b. The increase in the activity of the supercomplexes in comparison with the individual complex IV was as follows: in bovine, pig, wild boar, deer and mouse heart mitochondria for supercomplex b an 8.3-fold increase and for supercomplex c 8.0-fold (except the mouse samples). In rat heart mitochondria I1III2IV1 (b) is 11.6-fold and I1III2IV2 (c) 14.02-fold more active. The solubilisate of deer kidney mitochondria showed a smaller enhancement of activity for supercomplex b (factor 4.2) and c (factor 4.8). With antibodies against the complexes I, III, IV and V the individual complexes and higher assemblies were detected in 1-D Western blots of blue native gels. In accord with the in-gel activity tests for OxPhos complexes I and IV it was demonstrated that the signal of supercomplexes containing complex I was much stronger than that of the individual complexes. For complexes III2 and IV this was not observed, the signal of the individual complexes were more pronounced. For complex V (the ATP synthase) the momomeric and dimeric form as well as higher oligomers were found in all samples. The solubilisates of mouse, pig, deer, wild boar heart, and deer kidney were seperated by 2 D BN/SDS-PAGE and the mitochondrial proteins were identified by peptide mass fingerprint (MALDI-TOF MS). Because similar experiments were already done for bovine heart mitochondria by Kratochwil and Hunzinger et al. and rat heart mitochondria by Reifschneider et al. further analysis of these samples was omitted. Also Groebe et al. examined the mitochondrial proteins in human and Podospora anserina samples. In this thesis between 29 and 36 proteins, dependent on the tissue sample, were identified. Approximately 80 % of these proteins were subunits of the OxPhos complexes I, III, IV and IV. It was demonstrated that a large part of the identified subunits possessed a similar electrophoretic mobility. A comparison with the results of bovine and rat heart mitochondria showed a high consistency. Especially for the ATP synthase (complex V) in the examined tissues subunits for the monomeric, dimeric and higher oligomeric forms were found. Additional subunits of the OxPhos complexes were identified by 2-D Western blots. At these conditions (2-D BN/SDS-PAGE) complex II was not involved in formation of supercomplexes and existed only as individual complex. Quantitative analysis of the identified protein spots was done by staining the 2-D gels with the fluorescent dye Sypro® Ruby. For data evaluation the software Delta2D was used. A comparison of bovine, pig, wild boar and deer heart mitochondria revealed that the protein amount for the OxPhos complexes I, III and V was quite similar. Only small differences in the ratio between supercomplexes and individual complexes were observed. In addition to these results it was demonstrated that the ATP synthase exits mostly in monomeric form, between 61 and 72 % dependent on the examined tissue sample. The largest difference was noted for the protein amount of complex IV. A comparison of bovine, mouse and rat heart mitochondria showed distinct differences for the protein amount in complex I and V. In rat heart samples the amount for the monomeric and dimeric form of the ATP synthase were quite similar in contrast to the other tissue samples. In mouse heart samples complex V was mostly in monomeric form (78 %). Furthermore, complex I was mostly assembled in supercomplex a. As for the other samples (bovine, rat, pig wild boar, deer) supercomplex b was always preferred. For the dimers and other oligomers of complex V is known that they are invovled in the formation of the cristae morphology in mitochondria. It is so far still unknown, if and how much the enzymatic activity is influenced by this oligomerization. Therefore the ATP hydrolysis activity of the monomer as compared to the oligomeric state was analyzed by in-gel activity tests. Initial experiments showed that the BN-PAGE was not suitable for this approach. The anionic dye Coomassie Brillant Blue interfered with the accessibility of the substrate and the detection of the preticipate on the gel. Therefore the milder colorless native electrophoresis (CN-PAGE) was used for further experiments. It was demonstrated for the first time that ATP synthase in deer, pig and wild boar mitochondria forms also oligomers, which exhibited a ATP hydrolysis activity. Similar results were found for the well examined rat, mouse and bovine heart samples. In contrast to the findings in the literature the monomeric form of complex V showed the highest hydrolysis activity, the dimers and other oligomers exhibited reduced activity depending on the degree of assembling. The relation between the hydrolysis and the synthesis of ATP by complex V is still unknown. If it is assumed that the reduction in the hydrolysis activity correlates to an increase in synthesis activity, then the synthesis rate would be increased with the oligomeric state of the ATP synthase. According to this the enzymatic activity could then be specifically adjusted to the demand of the cell without changing the de novo expression rate of complex V. The research of Meyer zu Tittingdorf et al. and Hema et al. demonstrated that environmental stress factors affect the metabolic state in mitochondria of the green algae Chlamydomonas reinhardtii. Therefore the influence of different temperatures during the cultivation period on the organization and the enzymatic activity of the OxPhos complexes was analyzed in this thesis. Two cultivation conditions were examined: In photomixotrophic growth the medium contained acetate as a additional carbon source, in photoautotrophic growth carbon dioxide from the air was assimilated. For a functional analysis of the OxPhos complexes I and IV the described in-gel activity tests were employed. Similar to mitochondria in mammals the complexes I, III2 and V of C. reinhardtii assemble into supercomplexes, I1III2IV1 (S1) and I1III2IV2 (S2). Under photomixotrophic conditions an elevation of temperature during the cultivation period led to an increase in the activity of complex I (1.6-fold) and IV (1.8-fold) in the supercomplexes S1 and S2. The individual complex showed no increase in activity. Similar tendencies were observed for complex I under photoautotrophic cultivation. In contrast to these findings the activity of complex IV exhibited a small decrease while cultivated at higher temperatures. A comparison of the activity of the individual complexes with the supercomplexes S1 and S2 revealed that the assembling into supercomplexes in plant-like organisms led to an increase in activity. For complex I the enhancement was 5-fold and for complex IV 10-fold. The results of this thesis can be summarized as follows: In mitochondria from mammalian heart tissues and from the green algae C. reinhardtii the OxPhos complexes I, III2 and IV are assembled into supercomplexes. In all examined samples the supercomplexes showed a significant increase in enzymatic activity in comparison to the individual complexes. For the ATP synthase it was demonstrated that the ATP hydrolysis activity was influenced by the oligomeric state. In coorporation with the research groups of Prof. Dr. Grüber (NTU, Singapore) and of Prof. Dr. Gordeliy (IBS Grenoble; ICS Jülich; MIPT Moscow) the structure of the chloroplast ATP synthase was examined by X-ray crystallography. Crystal structures of the hydrophilic domain F1 and some isolated subunits of the membran integral FO-domain are known, but to date nobody was able to crystallize the whole intact enzyme complex. The chloroplast ATP synthase from spinach (Spinacia oleracea) was isolated in sufficient amount (100-200 mg). By BN-PAGE it was demonstrated that the enzyme complex was intact after the isolation. For measuring the synthesis activity, the ATP synthase was reconstituted in liposomes, an electrochemical proton gradient (∆Ψ+∆pH) was applied and subsequently an luciferin-luciferase-ATP-assay was performed. In Prof. Dr. Grüber´s research group Dr. Asha Manikkoth Balakrishna used the vapour diffusion method for crystallization experiments. The obtained crystals were analyzed by a synchrotron (Swiss Light Source) at the Paul Scherrer Institut (Switzerland). Except the c14-cylinder all other subunits were lost during the crystallization process, the calculated crystal structure had a resolution of 4,50 Å. In Prof. Dr. Gordeliy´s research group crystallization was done using the lipidic cubic phase method. While starting with the intact enzyme complex, under these conditions also only crystals of the c14-cylinder were obtained, but for the first time with a resolution of 2,48 Å.
