Abstract:

Die DHAP-Aldolasen stellen durch ihre relativ breite Substrattoleranz und ihre hohe Stereospezifität ein sehr interessantes Werkzeug dar, um neue komplexe Naturstoff-Analoga herzustellen. Die Synthese von komplexen Oligosaccharid-Mimetika wurde am Beispiel von zwei Strategien untersucht. • Bei der verwendeten Tandem-Aldoladdition handelt es sich um eine bidirektionale, enantio- und diastereoselektive Kettenverlängerung zu neuen komplexen Saccharid-Mimetika. Die Umsetzung von symmetrischen Diketonen lieferte nach einer Barbier-Allylierung, einer Ozonolyse, einer enzymatischen Aldoladdition und einer anschließenden Dephosphorylierung neue Spiro-Saccharide. Trotz der begrenzten Substrattoleranz wurden Edukte mit verschiedenen Symmetriemustern (C2, C2v) erfolgreich eingesetzt. • Strukturell modifizierte Analoga des Tumorantibiotikums Pancratistatin stellen potentielle neue Wirkstoffe dar. Die Struktur von Pancratistatin enthält einen polyhydroxylierten Cyclus, der durch einen Zuckerrest ersetzt werden kann. Aldolasen wurden für eine Kettenverlängerung auf dem Weg zum Endprodukt eingesetzt.

Trough their large substrate spectrum and their high stereoselectivity are DHAP-Aldolases a powerful tool for the synthesis of new carbohydrate mimetics as potential biologically active compounds. Two methodologies were studied. • Though the tandem aldol addition, a bi-directional, enantio- and diastereoselective chain elongation, new spiro-saccharides were prepared from diketones with different symmetry patterns, C2, C2v. • A stereospecific synthesis of novel pancratistatin analogues were presented. The study demonstrates that structures of high molecular complexity, incorporating a manifold of contiguous chiral centers, can be readily prepared in a few synthetic steps by a combination of enzymatic dihydroxylation and aldolization with DAHP-aldolases.
