Abstract:

Die bisher einzig bekannte Totalsynthese von Fast Blue basiert auf den Arbeiten von Otto Dann im Rahmen seiner Leukämieforschung Anfang der 80er Jahre des letzten Jahrhunderts. Ihm gelang der Aufbau von Fast Blue über 15 Stufen bei einer Gesamtausbeute von 1.5% (76% pro Stufe). Die Entdeckung weiterer pharmakologisch relevanter Eigenschaften durch die Gruppe um Lazo et al., wie die Protein-Kinase-Phosphatase Inhibierung, zeigt deutlich die Relevanz und Aktualität von Fast Blue. Die vorliegende Arbeit beschreibt die Entwicklung eines neuen synthetischen Zugangs zu Fast Blue, der neben einer reduzierten Stufenzahl (und damit auch eine Erhöhung der Gesamtausbeute) auch eine ausgeprägte strukturelle Vielfalt im Hinblick auf eine spätere Derivatisierung ermöglichen soll. Es wurden acht verschiedene Routen zur Synthese von FB entwickelt und untersucht, welche nach dem jeweiligen Schlüsselschritt der Sequenz benannt wurden. Die erprobten Synthesestrategien umfassen die Julia-Kocienski-Route, die „oxidative“ sowie die „klassische“ Heck-Route. Auf Basis der „oxidativen“ Heck-Reaktion werden neue Vorgehensweisen untersucht, darunter die Verwendung der 2-(Pyridylsulfonyl)-Gruppe als dirigierende Schutzgruppe, eine „oxidative“ Kupplung nach Miura et al. sowie eine „umgepolte“ Variante. Als Weiterentwicklung der „klassischen“ Heck-Variante wurde eine Tandem-Variante unter Verwendung des trans-Brom-iodethens erprobt. Abschließend wurden zwei Syntheserouten vorgestellt, welche den Aufbau des Indolkörpers als zentrales Syntheseelement beinhalten. Dabei handelt es sich um eine intramolekulare Variante der Heck-Reaktion sowie die Amid-Wittig-Route.

Erst im Rahmen der Amid-Wittig-Route konnte Fast Blue über insgesamt 11 Stufen mit einer Gesamtausbeute von 5.4% hergestellt werden. Im Vergleich zur Dann`schen Syntheseroute erfolgt eine Steigerung um den Faktor 3.6, bei Einsparung von vier Syntheseschritten.

Fast Blue was exclusively synthesized by Otto Dann during his leukemia research program at the beginning of the 1980s. He was able to build up Fast Blue over the course of 15 steps with an overall yield of 1.5% (76% per step). The discovery of new relevant pharmacological features like the protein-kinase-phosphatase inhibition by the group of John Lazo et al. symbolizes the importance and actuality of Fast Blue. This thesis deals with the development of a new synthetic approach to Fast Blue. Decrease of total steps count (including the enhancement of overall yield) and further easy structural diversification are major tasks of the development. Eight new synthetic routes (which are named after the key step) were created and evaluated. These strategies contain the Julia-Kocienski-route, the „oxidative“ and „classical“ Heck-routes. Based on the “oxidative” Heck reaction new approaches were evaluated, including the 2-(Pyridylsulfonyl)-group as a direction protection group, the “oxidative” coupling presented by Miura et al. and a so-called “umgepolte” variant. As an advancement of the prototype of the “classical” Heck reaction a tandem-modification by usage of trans-1-Bromo-2-iodoethene was evaluated. Initial success was achieved during the Amid-Wittig sequence. Here Fast Blue was synthesized over the course of 11 steps with an overall yield of 5.4%. In comparison to Danns` work it is a percentage increase of 260% by saving four steps.
