Abstract:

The availability of fast calculation methods for molecular thermodynamics is the basis for the efficient computational treatment of complexation phenomena in solution. Free energies of binding are often determined by statistical sampling of the intermolecular free energy surface. Detailed molecular dynamics simulation and Monte-Carlo-simulation of complex systems on the atomic level has become feasible, but suffers from the large computational effort spent in sampling the solvent configurational space. However, the calculation times can be reduced by using combinations of simulation and integral equation methods. The focus of this work is the development of hybrid simulation/integral equation methods and their application to the calculation of thermodynamic and structural properties of crown ether complexes in solution. Therein, the solvent is modeled by an integral equation representation based on radial distribution functions. Three methods are presented, accounting for thermodynamic simulation, local and global optimization of complex structures. The hybrid Monte-Carlo/RISM approach is well suited to explore complexation free energy hypersurfaces in solution. Using MC/RISM, studies are presented concerning the complexation thermodynamics of metal ions, organic ammonium ions and guanidinium derivatives with crown ethers in water, methanol and acetonitril. Local optimization techniques are used to calculate and visualize local properties of free energy surfaces. Complexes of chiral crown ethers and racemic organic ammonium ions are known to exhibit stereoselective differentiation mechanisms which depend on the nature of the solvent. This phenomenon is studied by means of molecular docking. A genetic algorithm is combined with RISM to perform docking studies of stereoisomeric complex partners in methanol, resulting the chiral selection in terms of structural and energetic information.

Die schnelle, computergestützte Berechnung thermodynamischer Eigenschaften ist die Basis zur in-silico-Untersuchung supramolekularer Verbindungen in Lösung. So können Freie Bindungsenthalpien durch statistisches Sampling der intermolekularen Freien Energiefläche bestimmt werden. Diese Technik ist im Rahmen der Molekulardynamik und Monte-Carlo-Methodik bereits etabliert, jedoch stets mit einem hohen numerischen Aufwand verbunden, der auch auf die explizite Behandlung von Solvensmolekülen zurückgeht. Durch Kombination von Simulations- und Integralgleichungsmethoden kann dieser Aufwand maßgeblich reduziert werden. Im Rahmen der vorliegenden Arbeit werden hybride Simulations-/Integralgleichungsmethoden entwickelt und ihre Eignung zur Berechnung struktureller und thermodynamischer Eigenschaften von Kronenetherkomplexen in Lösung gezeigt. Das Lösungsmittel wird dabei jeweils durch radiale Verteilungsfunktionen auf der Basis der RISM-Integralgleichungsmethode modelliert. Im einzelnen werden Verfahren zur lokalen und globalen Komplexstruktur-Optimierung und zur thermodynamischen Simulation vorgestellt. Die Kombination von Monte-Carlo-Simulation mit RISM kann zur schnellen Berechnung Freier Energieflächen in Lösung dienen. Auf der Basis dieses Verfahrens wird die Komplexbildung von Kronenethern mit Alkalimetallen und molekularen Ammoniumionen in den Lösungsmitteln Wasser, Methanol und Acetonitril untersucht. Dabei finden auch lokale Aspekte der Energiehyperflächen in diesen Systemen Erwähnung. Beim Einbringen chiraler Gruppen in das beschriebene System Kronenether-Ammoniumion kann oft eine lösungsmittelabhängige chirale Differenzierung beobachtet werden. Zur Berechnung dieser differenziellen Größe wird RISM mit einem genetischen Algorithmus und einem lokalen Optimierer verknüpft. Auf diese Weise gelingt die Berechnung der chiralen Selektion stereoisomerer Komplexpaare im Lösungsmittel Methanol.
