Abstract:

Die Kopplung zwischen Protonenpumpe und Protonenverbraucher durch einen elektrochemischen Protonengradienten stellt ein ubiquitäres Motiv in der belebten Natur dar. Nach wie vor ist umstritten, ob die gepumpten Protonen direkt entlang der Membran von der Protonenpumpe zum Verbraucher diffundieren oder ob zunächst eine Equilibrierung mit der umgebenden wäßrigen Phase stattfindet. Die Protonentranslokation läßt sich mit Hilfe der zeitaufgelösten Blitzlicht-Spektroskopie an Modellsystemen untersuchen. Als Protonenpumpe wurde dabei Bacteriorhodopsin in Form von Purpurmembran und rekonstituiert in Proteoliposomen eingesetzt. Die CFoF1-ATP-Synthase aus Spinacia oleracea diente in diesem System als Protonenverbraucher. Die Protonentranslokation kann in einem solchen System anhand von Absorptionsänderungen eingebrachter pH-Indikatoren detektiert werden. Durch spezifische Kopplung von pH-Indikatoren mit den Proteinen im Modellsystem kann sowohl die Kinetik der Protonenfreisetzung bestimmt werden, als auch die Diffusion entlang der Membran und der Austausch mit der umgebenden wäßrigen Phase. Die Untersuchungen haben gezeigt, daß der Diffusionskoeffizient für Protonen entlang der Purpurmembran etwa 60fach kleiner ist als für Protonen in der wäßrigen Phase. Trotzdem sind die Protonen auf der zytoplasmatischen Seite der Membran deutlich früher zu detektieren als in der umgebenden wäßrigen Phase. Die Untersuchung der Protonentranslokation an rekonstituierten Proteoliposomen zeigte, daß hier der Austausch von Protonen mit der wäßrigen Phase gegenüber der Purpurmembran deutlich beschleunigt ist. Eine Abhängigkeit dieser Kinetik von der Zusammensetzung der Lipidphase und insbesondere vom Ladungszustand der Lipide konnte jedoch nicht gefunden werden.

The energetic coupling between a proton pump and a proton sink via an electrochemical proton gradient across a lipid membrane is an universal motif in biology. Primary available sources of energy are converted into the biological usable energy of ATP. For the investigation of this coupling, we use a model system, consisting of the light-driven proton pump bacteriorhodopsin from Halobacterium salinarum and the CFOF1-ATP-synthase of spinach. It is still disputed, if the protons migrate directly along the membrane from the source to the sink or if the protons first exchange with the surrounding water and diffuse through the water. As protons cannot be detected directly, optical pH-indicators as probe must be inserted into the system. The proton movement can then be obtained as absorption changes of the indicators. The negatively charged indicator pyranine is preferred for the aqueous phase. Various reactive derivatives of fluorescein can specifically be linked to different sites of BR and the ATP-synthase. With these indicators at the different sites of the system we get a temporal and spatial resolution of the proton migration. The investigations showed, that lateral proton migration on the membrane is 60 times slower than proton diffusion in bulk water. But a dependence of the kinetic on the composition of the lipid phase and especially on the charge of the lipids could not be found.
