Abstract:

Ziel dieser Arbeit war die Konstruktion und Anwendung eines auf GFP basierenden Reportersystems und eines induzierbaren Genexpressionssystems in Haloarchaea. Die Anwendung von Reporter- oder Expressionssystemen in Haloarchaea ist aufgrund deren hohen, intrazellulären Salzkonzentration limitiert. Somit stehen zur Untersuchung der Regulation der haloarchaealen Genexpression nur wenige solcher Systeme zur Verfügung. Daher wurden bakterielle wie auch eukaryotische Komponenten zum Aufbau eines Reporter- sowie eines induzierbaren Genexpressionssystems in Haloarchaea verwendet. Im ersten Teil dieser Arbeit wurde ein auf GFP basierendes Reportersystem konstruiert. Ausgangspunkt war die unter hypersalinen Bedingungen funktionsfähige GFP-Variante smRS-GFP, die sich jedoch zur Quantifizierung schwacher Promotoren als nicht geeignet erwies. Durch die Insertion von insgesamt zehn Aminosäuresubstitutionen in smRS-GFP wurde mGFP6 mit einer 3,3-fach stärkeren Fluoreszenz generiert. Mittels mGFP6 konnten sechs haloarchaeale Promotoren unterschiedlicher Stärke charakterisiert werden. Die höchste Aktivität wiesen die beiden housekeeping-Promotoren Pfdx des Ferredoxin-Gens und P2 der ribosomalen 16S rRNA auf. Deutlich geringere Aktivitäten sowie ein wachstumsabhängiges Aktivitätsmuster wurden bei den Promotoren PpA, PpO, PpD und PpF der p-vac-Region festgestellt, die die Expression von Gas-Vesikel-Protein-Genen (p-gvp) in Halobacterium salinarum PHH1 kontrollieren. Auch zeigte sich eine Regulation ausgehend der 5′-UTR sowie der 12 nt ab Position +1 verschiedener p-gvp-mRNAs. Eine Deletion der 5′-untranslatierten Region (5′-UTR) oder der 12 nt stromabwärts des AUG-Startcodons resultierte immer in eine Erhöhung der Expression. Die Promotoren PpA und PpD werden von dem Aktivatorprotein pGvpE induziert. Mit mGFP6 wurden eine bis zu 13-fache Aktivierung von PpA und eine bis zu 8-fache Aktvierung von PpD durch GvpE festgestellt. Zudem wurde mit mGFP6 ein Einfluss der 5′-UTR des p-gvpD-Gens auf die GvpE-vermittelte Aktivierung von PpA und PpD festgestellt. Von den mittels mGFP6 charakterisierten haloarchaealen Promotoren und Regulatorelementen eigneten sich besonders die beiden starken Promotoren Pfdx und P2 zum Aufbau eines induzierbaren Genexpressionssystems, das im zweiten Teil dieser Arbeit generiert wurde. Im Fokus der regulatorischen Elemente standen hier die synthetischen Theophyllin-abhängigen Riboswitche A bis E und E* (zur Regulation der Translation). Nur die Insertion des Theophyllin-Riboswitches E stromaufwärts des bgaH-Reportergens führte zu einer regulierbaren Expression mit einer 3-fachen Aktivierung bei Anwesenheit von Theophyllin. Zudem wurde eine Salz- sowie Temperaturabhängigkeit des Theophyllin-Riboswitches E nachgewiesen, was in einer 12-fachen (bei Erhöhung des Salzgehaltes) beziehungsweise in einer 26-fachen Aktivierung (bei Erniedrigung der Temperatur) der Translation der bgaH-mRNA resultierte. Eine vollständige Reprimierung der Expression in Abwesenheit von Theophyllin wurde nicht erreicht. Mit dem mGFP6-Reporter- und dem Theophyllin-abhängigen Genexpressionssystem wurde final ein Zusammenhang zwischen der GvpE-Menge und der Höhe der Aktivierung von PpA festgestellt. Somit stehen nun zwei weitere Werkzeuge zur Untersuchung der haloarchaealen Genexpression zur Verfügung, die zudem miteinander kombiniert werden können.

The aim of this work was to construct and apply a GFP-based reporter system and an inducible gene expression system in haloarchaea. The application of reporter or expression systems in haloarchaea is limited due to their high, intracellular salt concentration. Thus, few such systems are available to study the regulation of haloarchaeal gene expression. Therefore, bacterial as well as eukaryotic components were used to construct a reporter as well as an inducible gene expression system in haloarchaea. In the first part of this work, a GFP-based reporter system was constructed. The starting point was the GFP variant smRS-GFP, which is functional under hypersaline conditions, but proved to be unsuitable for quantifying weak promoters. Insertion of a total of ten amino acid substitutions into smRS-GFP generated mGFP6 with 3.3-fold stronger fluorescence. Using mGFP6, six haloarchaeal promoters of different strengths could be characterized. The two housekeeping promoters Pfdx of the ferredoxin gene and P2 of the ribosomal 16S rRNA showed the highest activity. Significantly lower activities as well as a growth-dependent activity pattern were observed for the promoters PpA, PpO, PpD, and PpF of the p-vac region, which control the expression of gas vesicle protein genes (p-gvp) in Halobacterium salinarum PHH1. Also, regulation was shown starting from the 5′-UTR as well as the 12 nt from position +1 of different p-gvp mRNAs. Deletion of the 5′-untranslated region (5′-UTR) or the 12 nt downstream of the AUG start codon always resulted in an increase in expression. The promoters PpA and PpD are induced by the activator protein pGvpE. With mGFP6, up to 13-fold activation of PpA and up to 8-fold activation of PpD by GvpE were detected. In addition, with mGFP6, an effect of the 5′-UTR of the p-gvpD gene on GvpE-mediated activation of PpA and PpD was detected. Of the haloarchaeal promoters and regulatory elements characterized using mGFP6, the two strong promoters Pfdx and P2 were particularly suitable for building an inducible gene expression system, which was generated in the second part of this work. The regulatory elements here focused on the synthetic theophylline-dependent riboswitches A to E and E* (for regulation of translation). Only insertion of the theophylline riboswitch E upstream of the bgaH reporter gene resulted in regulatable expression with 3-fold activation in the presence of theophylline. In addition, a salt- as well as temperature-dependence of theophylline riboswitch E was demonstrated, resulting in a 12-fold (when salinity was increased) and a 26-fold (when temperature was decreased) activation of translation of bgaH mRNA, respectively. Complete repression of expression in the absence of theophylline was not achieved. Using the mGFP6 reporter and theophylline-dependent gene expression systems, a correlation between the amount of GvpE and the level of activation of PpA was finally established. Thus, two additional tools for the study of haloarchaeal gene expression are now available and, moreover, can be combined.
