Abstract:

Polyaspartate sind bekannt für die Ausbildung vielfältiger, teils geordneter Sekundärstrukturen. In dieser Dissertation wird die Synthese und Untersuchung stimuliresponsiver Polyaspartate beschrieben, welche in vier Abschnitte (Projekte) unterteilt ist. Für Projekt A wird Poly-β-benzyl-ʟ-aspartat (PBLA) als neues Orientierungsmedium für die NMR-Spektroskopie etabliert und dessen Eigenschaften mit dem bekannten Orientierungsmedium auf Basis von Poly-γ-benzyl-ʟ-glutamat (PBLG) verglichen. In Projekt B werden zwei helikale Polypeptide (Poly-β-benzyl-ʟ-aspartat-co-poly-β-phenethyl-ʟ-aspartat, PBLA-co-PPLA und Poly-γ-p-biphenylmethyl-ʟ-glutamat, PBPMLG) selektiv deuteriert und deren Deuterium-Quadrupolaufspaltung mittels NMR-Spektroskopie gemessen. Auf diese Weise werden deren thermoresponsive Eigenschaften (Helixinversion und Helixreorientierung) untersucht. In Projekt C wird eine Methode zur Messung des Circulardichroismus anisotroper Proben entwickelt und überprüft. Im Folgenden wird die Methode zur weiteren Untersuchung von PBLA-co-PPLA genutzt. In Projekt D wird abschließend eine neue Klasse photoresponsiver (Co-) Polyaspartate auf Basis von ortho-fluorierten Azobenzolen beschrieben. Diese Polymere zeichnen sich durch eine enorme Bandbreite an photoschaltbaren Sekundärstrukturen aus und werden hinsichtlich ihrer Fähigkeit zur Ausbildung anisotroper Phasen untersucht.

Polyaspartates are known to form a variety of (ordered) secondary structures. The synthesis and characterization of stimuliresponsive polyaspartates is the topic of this work, which is subdivided into four parts (projects). For project A poly-β-benzyl-ʟ-aspartate (PBLA) is developed as a new alignment medium for the use in NMR-spectroscopy and is compared with poly-γ-benzyl-ʟ-glutamate (PBLG), which has been a well-known alignment medium already. In project B two helical polypeptides (poly-β-benzyl-ʟ-aspartate-co-poly-β-phenethyl-ʟ-aspartate, PBLA-co-PPLA and poly-γ-p-biphenylmethyl-ʟ-glutamate, PBPMLG) are specifically deuterated and their deuterium quadrupolar splitting is measured by means of NMR-spectroscopy. Based on these measurements, their thermoresponsive behaviours (helix inversion and helix realignment) were observed for the first time and investigated in more detail, respectively. In project C a method for measurement of the circular dichroism of anisotropic samples is developed and evaluated. The method is further used for investigation of PBLA-co-PPLA. In project D a new class of photoresponsive (co-) polyaspartates on the basis of ortho-fluorinated azobenzenes is described. These polymers exist in a variety of photoresponsive secondary structures and are investigated with respect to their ability to form anisotropic phases.
