Abstract:

Toluol wird unter anaeroben Bedingungen durch das denitrifizierende Betaproteobakterium T. aromatica abgebaut. Die initiale Reaktion des Stoffwechsels ist die Addition der Methylgruppe von Toluol an die Doppelbindung von Fumarat. Dieser Schritt, bei dem (R)-Benzylsuccinat entsteht, wird durch die (R)-Benzylsuccinat-Synthase katalysiert. Dieses Enzym gehört zur Familie der Glycylradikal-Enzyme und wird erst posttranslational durch ein aktivierendes Enzym zur aktiven, Glycylradikal-haltigen Form umgesetzt. (R)-Benzylsuccinat wird anschließend über eine β-Oxidations-ähnliche Kaskade von Enzymen zu Benzoyl-CoA und Succinyl-CoA umgesetzt. Benzoyl-CoA ist ein zentrales Intermediat des anaeroben Stoffwechsels, dessen weiterer Abbauweg bereits gut bekannt ist. Ziel dieser Arbeit war die Charakterisierung der letzten drei Enzyme dieses Weges, welche die Umsetzung von (E)-Phenylitaconyl-CoA zu Benzoyl-CoA und Succinyl-CoA katalysieren. Des Weiteren wurde die Aktivierbarkeit der rekombinanten (R)-Benzylsuccinat-Synthase durch das aktivierende Enzym überprüft. 1. Die (E)-Phenylitaconyl-CoA Hydratase wurde heterolog in E. coli überproduziert, gereinigt und charakterisiert. Es handelt sich um ein Homotetramer aus 29 kDa-Untereinheiten und das Produkt der Reaktion wurde über HPLC-MS als 2-(Hydroxymethylphenyl)-Succinyl-CoA identifiziert. Das Enzym zeigt eine kooperative Kinetik mit einem Hill-Koeffizienten von N=4, einem apparenten Km-Wert von 109 ± 7 µM und Vmax von 32 ± 3 µmol min-1 (mg Protein)-1. Des Weiteren unterliegt das Enzym einer Substratinhibition (Kis: 234 ±12 µM). Mit einem alternativen Substrat, das statt mit CoA durch N-Acetylcysteamin aktiviert war, wurden vergleichbare enzymkinetische Parameter erhalten. 2. Die 2-(Hydroxymethylphenyl)-Succinyl-CoA-Dehydrogenase wurde in E. coli überproduziert, gereinigt und näher charakterisiert. In einem gekoppelten Enzymtest folgt das heterodimere Enzym einer Michaelis-Menten-Kinetik mit einem apparenten Km-Wert von 84 ± 8 µM und einem Vmax von 1,2 ± 0,1 µmol min-1 (mg Protein)-1. Das Produkt der Reaktion wurde über RP-HPLC detektiert und die im Produkt vorhandene Ketogruppe durch Bildung eines Hydrazons nachgewiesen. In der Rückreaktion reduziert das Enzym Halogen-substituierte Acetophenone mit einer kooperativen Kinetik. 3. Die Thiolase-Untereinheit BbsB wurde zusammen mit dem konservierten BbsA-Protein in E. coli überproduziert. Die beiden Proteine wurden während der Reinigung getrennt und separat charakterisiert. BbsB und BbsA bilden jeweils ein Homodimer aus 41 kDa- bzw. 16 kDa-Untereinheiten, das BbsA-Protein enthält zudem ein Zink-Ion pro Untereinheit, welches vermutlich über ein konserviertes Zinkfinger-Motiv der Sequenz gebunden ist. Die erwartete Aktivität der Thiolase wurde über RP-HPLC in Form der Rückreaktion als Kondensation von Succinyl-CoA und Benzoyl-CoA gemessen und war strikt abhängig von der Anwesenheit beider Proteine BbsA und BbsB. Die Ketogruppe im Reaktionsprodukt wurde durch Derivatisierung mit Phenylhydrazin nachgewiesen, und über einen photometrischen Enzymtest wurde der Reaktionsmechanismus der Benzoylsuccinyl-CoA-Thiolase als Ping-Pong-Kinetik mit den folgenden Parametern bestimmt: apparente Km-Werte von 155 ± 12 µM für Benzoyl-CoA bzw. 190 ± 6 µM für Succinyl-CoA und ein Vmax-Wert von 11 ± 2 µmol min-1 (mg Protein)-1. 4. Durch Optimierung der Anzucht- und Aufarbeitungsbedingungen von T. aromatica bzw. Azoarcus sp. Stamm T wurden Rohextrakte mit spezifischen Aktivitäten beider (R)-Benzylsuccinat-Synthasen von 18 ± 2 µmol min-1 (mg Protein)-1 bzw. 9 ± 1,6 µmol min-1 (mg Protein)-1 erhalten. In Rohextrakten beider Stämme wurde zusätzlich die Umsetzung eines weiteren Substratanalogs (o-Toluidin) nachgewiesen, das mit 9-13 % der Rate der Toluol-Umsetzung zum Succinataddukt umgesetzt wurde. 5. Das aktivierende Enzym wurde als Fusionsprotein in E. coli überproduziert und lag zu 10 % in löslicher Form vor. Aktivierungsversuche mit gereinigter rekombinanter (R)-Benzylsuccinat-Synthase durch Zellextrakte mit überproduziertem aktivierendem Enzym und zugesetzten inaktivierten Rohextrakten von T. aromatica bzw. Azoarcus sp. Stamm T lieferten allerdings weder über Messung produzierten (R)-Benzylsuccinats noch über Western-Blot-Analyse der radikaltragenden Untereinheit Hinweise für die erwartete Glycyl-Radikal-Bildung. Die Gene bssCAB, die für die (R)-Benzylsuccinat-Synthase in G. metallireducens kodieren, wurden in einen Überexpressionsvektor kloniert. Jedoch wurde keine Überproduktion des Proteins in E. coli beobachtet. 6. Die vermuteten akzessorischen Genprodukte BssE, BssF und BssG wurden in E. coli überproduziert, liegen aber fast ausschließlich in Form von Einschlusskörperchen vor.

Toluene is degraded under anaerobic conditions by the denitrifying beta-proteobacterium T. aromatica. The initial reaction of anaerobic catabolism of toluene is the addition of the methyl group of toluene on the double bond of fumarate. This step is catalysed by the enzyme (R)-benzylsuccinate synthase and yields(R)-benzylsuccinate. The enzyme belongs to the family of glycyl radical enzymes and an activating enzyme introduces the radical posttranslationally into (R)-benzylsuccinate synthase. Further pathway of anaerobic toluene catabolism follows a modified ß-oxidation of (R)-benzylsuccinate to benzoyl-CoA, the common intermediate in aromatic metabolism, and succinyl-CoA. The main topic of this work was characterisation of three enzymes which are assumed to be involved in β-oxidation of (E)-phenylitaconyl-CoA to benzoyl-CoA and succinyl-CoA and had not been investigated prior to the work. Also activation of (R)-benzylsuccinate synthase by the activating enzyme was verified. 1. (E)- phenylitaconyl-CoA hydratase was overproduced in E. coli, purified and characterised. The composition of the enzyme was identified as a homoteramer composed of subunits of 28 kDa. Product of reaction was identified by HPLC-MS as 2-(hydroxymethylphenyl)-succinyl-CoA. The enzyme shows cooperativity with a hill-coefficient of N = 4, an apparent Km-value of 109 ± 7 µM and a Vmax of 32 ± 3 µmol min-1 (mg protein)-1. Also substrate inhibition was observed (Kis: 234 ± 12 µM). 2. 2-(hydroxymethylphenyl)-succinyl-CoA dehydrogenase was overproduced in E. coli, purified and characterised. In a coupled enzyme assay the enzyme follows a kinetic of michaelis-menten with an apparent Km-value of 84 ± 8 µM and a Vmax von 1.2 ± 0.1 µmol min-1 (mg protein)-1. The reaction product was detected by RP-HPLC and in presence of phenylhydrazine the ketogroup in this product was verified. The enzyme reduces halogen-substituted acetophenones in back reaction with a cooperative kinetic. 3. Thiolase subunit BbsB and the conserved subunit BbsA were overproduced in E. coli. Both proteins could be separated of each other and characterised. Both enzymes were identified as homodimers of 41 kDa- and 16 kDa-subunits respectively. BbsA also contains a zinc-ion per subunit which is probably bound by a conserved zinc-finger-motiv. Activity as thiolase was measured by product formation of condensation of benzoyl-CoA and succinyl-CoA via RP-HPLC and was only detectable in presence of BbsA and BbsB. The ketogroup of the reaction product was verificated by derivatisation with phenylhydrazine. By a photometric enzyme assay the reaction mechanism of benzoylsuccinyl-CoA-thiolase was determined as ping-pong kinetic with following parameters: apparent Km-value of 155 ± 12 µM for benzoyl-CoA, 190 ± 6 µM for succinyl-CoA and a Vmax-value of 11 ± 2 µmol min-1 (mg protein)-1. 4. Improvement of cultivation- and preparation-conditions of T. aromatica and Azoarcus sp. strain T allows measurements of specific activities of (R)-benzylsuccinate-synthases of 18 ± 2 µmol min-1 (mg protein)-1 and 9 ± 1.6 µmol min-1 (mg protein)-1. Additionally conversion of a substrate ortholog (o-toluidine) in both cell extracts with 9 - 13 % of toluene conversion was determined. 5. Activating enzyme was produced toward 10 % as soluble fusion protein in E. coli. Activation of purified recombinant (R)-benzylsuccinate-synthase by activating enzyme and inactivated cell extracts of T. aromatica and Azoarcus sp. Strain T provides no evidence for accumulation of (R)-benzylsuccinate. Furthermore western-blot analyses of radical-containing subunit indicate no glycyl-radical-formation. bbsCAB of G. metallireducens were cloned in a overproduction vector, but no production in E. coli was observed. 6. The assumed accessory gene products BssE, BssF and BssG were overproduced exclusively as inclusion bodies in E. coli.
