Abstract:

Die D-Fructose-6-phosphat-Aldolase (FSA) und die D-Desoxy-5-phosphat-Aldolase (DERA) wurden als geeignete Biokatalysatoren für die asymmetrische Synthese von chiralen Molekülen in nicht-natürlichen Reaktionen untersucht. Das Donor- und Akzeptorspektrum der FSA wurde relativ breit studiert, wobei bisher nur einfache, sterisch weniger anspruchsvolle Aldol-Akzeptoren eingesetzt wurden. Im ersten Teil dieser Arbeit wurde untersucht, inwieweit dieses Spektrum auf sterisch anspruchsvollere Substrate ausgedehnt werden kann. Der Schwerpunkt lag dabei auf C-C-Bindungsreaktionen mit 3-Hydroxyaldehyden und Dialdehyden. Diese Ausgangssubstrate wurden mit Hilfe von FSA-Varianten mit Hydroxyaceton oder Dihydroxyaceton zu entsprechenden Desoxyzuckern umgesetzt und mittels NMR, HPLC und MS vollständig analysiert, wobei 21 neue Verbindungen erhalten wurden. Es wurde beobachtet, dass nur der Wildtyp der FSA gegenüber sterisch anspruchsvollen Substraten weitgehend tolerant war, während Tandemreaktionen nur bei linearen Dialdehyden funktionierten. Der zweite Teil der Arbeit befasste sich mit der Verwendung von DERA, seinen Varianten und verwandten Aldolasen für die Synthese von chiralen hydroxylierten C7/C8-Carbonsäuren ausgehend von Bernsteinsäure-Semialdehyd (BSA). Bislang gibt es keine ausreichend praktikable, kostengünstige Synthese für BSA. Daher wurde zunächst dessen Synthese für Aldolase-katalysierte Reaktionen etabliert, die in einer neuen zweistufigen Reaktionsfolge erfolgreich durchgeführt wurde. Die Umsetzung von BSA mit Acetaldehyd- und Acetonderivaten als Donorsubstrate ergab chirale hydroxylierte C7/C8-Carbonsäuren, die als Bausteine zur Erweiterung des Spektrums komplexer hydroxyfunktionalisierter Naturstoffe dienen können.

D-fructose-6-phosphate aldolase (FSA) and D-deoxy-5-phosphate aldolase (DERA) were investigated as suitable biocatalysts for the asymmetric synthesis of chiral molecules in non-natural reactions. The donor and acceptor spectrum of the FSA was studied relatively broadly, whereby only simple, sterically less demanding aldol acceptors were used so far. In the first part of this work we investigated to what extent this spectrum can be extended to sterically more demanding substrates. The focus was on C-C binding reactions with 3-hydroxyaldehydes and dialdehydes. These starting substrates were converted with hydroxyacetone or dihydroxyacetone to corresponding deoxysugars using FSA variants and completely analyzed by NMR, HPLC and MS, resulting in 21 new compounds. It was observed that only the wild type of FSA was largely tolerant to sterically demanding substrates, while tandem reactions only worked with linear dialdehydes. The second part of the work dealt with the use of DERA, its variants and related aldolases for the synthesis of chiral hydroxylated C7/C8-carboxylic acids starting from succinic acid semialdehyde (SSA). So far, there is no sufficiently practicable, cost-effective synthesis for SSA. Therefore, its synthesis for aldolase-catalyzed reactions was first established, which was successfully performed in a new two-step reaction sequence. The reaction of SSA with acetaldehyde and acetone derivatives as donor substrates yielded chiral hydroxylated C7/C8-carboxylic acids which can serve as building blocks for extending the spectrum of complex hydroxy-functionalized natural products.
